Aging, Cellular Senescence, and Alzheimer's Disease

Rui-Ming Liu

Summary: Cellular senescence plays a significant role in Alzheimer's disease pathophysiology, and targeting senescent cells may offer a promising approach for effective treatment of aging-related diseases like AD. However, the mechanisms promoting cell senescence and their contribution to AD neuropathophysiology remain largely unknown.

Identification and Validation of Aging-Related Genes in Idiopathic Pulmonary Fibrosis

Jie He et al.

Summary: In this study, 16 potential aging-related genes associated with idiopathic pulmonary fibrosis (IPF) were identified and validated using bioinformatics analysis and clinical sample validation. Among these genes, IGF1, RET, and IGFBP2 were found to play a role in the occurrence and prognosis of IPF. The findings may contribute to the understanding of IPF pathogenesis.

Differences Between Plasma and Cerebrospinal Fluid Glial Fibrillary Acidic Protein Levels Across the Alzheimer Disease Continuum

Andrea L. Benedet et al.

Summary: The study suggests that plasma GFAP is a sensitive biomarker for detecting reactive astrogliosis and Aβ pathology in early stages of AD, outperforming CSF GFAP. Plasma GFAP levels accurately discriminate between Aβ-positive and Aβ-negative individuals, with higher accuracy than CSF GFAP.

Genetic signature of human longevity in PKC and NF-κB signaling

Seungjin Ryu et al.

Summary: Gene variants associated with longevity were found to be enriched in the nPKC and NF-kappa B signaling pathways, with non-coding variants modulating the expression of cognate genes to reduce signaling. This matches genetic studies in other model organisms and suggests evolutionary conservation of reduced PKC and NF-kappa B signaling in exceptional longevity, possibly including humans.

Microglia Mediate the Occurrence and Development of Alzheimer's Disease Through Ligand-Receptor Axis Communication

Chongdong Jian et al.

Summary: This study conducted clustering analysis on single-cell RNA sequencing data of prefrontal cortex from 48 AD patients, identifying specific AD cell types and exploring the dysfunctional evolution of each cell type. The research also revealed the potential role of glial cells in the development of AD through cell communication, with key genes identified as potential biomarkers and therapeutic targets.

Discovery and Validation of Key Biomarkers Based on Immune Infiltrates in Alzheimer's Disease

Zhuohang Liu et al.

Summary: In AD patients, there are significant differences in immune cell infiltration, with higher proportions of monocytes, M0 macrophages, and dendritic cells compared to normal controls, while the proportions of other cells are lower. Furthermore, a model based on immune cell infiltration can be used to predict AD patient diagnosis and provide a new perspective for AD treatment targets.

Aberrant Methylation of Aging-Related Genes in Asthma

Yu Yang et al.

Summary: This study identified differential mRNA expression of aging-related genes in peripheral blood of asthma patients, as well as alterations in the methylation levels of these genes. Specific CpG sites were found to be associated with the severity of asthma, with nine CpG sites significantly correlated with pulmonary function parameters. ROC curve and PCA analysis indicated the potential use of these differential methylation sites as biomarkers for asthma.
