Efficacy and safety of esketamine nasal spray by sex in patients with treatment-resistant depression: findings from short-term randomized, controlled trials

Robyn R. Jones et al.

Summary: The objective of this analysis was to determine if there are sex differences with esketamine for treatment-resistant depression. Post hoc analyses of three randomized, controlled studies showed that esketamine was similarly effective and safe for both men and women with treatment-resistant depression.

Sex-specific effects of social isolation stress and ketamine on hippocampal plasticity

Jordan Logue et al.

Summary: Our study found that social isolation stress inhibits hippocampal LTP in male rats but does not affect PPF, an effect rescued by KET. In female rats, isolation stress does not alter LTP, but does reduce PPF - especially in diestrus - an effect rescued by KET at the highest dose. This suggests sex differences in the contribution of pre- and postsynaptic hippocampal compartments in response to stress and KET.

A systematic review of therapeutic ketamine use in children and adolescents with treatment-resistant mood disorders

Susan Kim et al.

Summary: Suicide is the second leading cause of death among individuals aged 10-24 in the United States, and ketamine has been shown to be effective in reducing depressive symptoms in adults. However, studies on ketamine use in youth are limited. Ketamine generally improves depressive symptoms, reduces acute suicidality, and mood lability in youth, although some individuals still show resistance to its treatment. Further longitudinal studies are needed to investigate ketamine's long-term safety, efficacy, and abuse potential in youth.

Summary: This study compared the effects of nortriptyline on adolescent and adult rats, finding that low doses had antidepressant effects in adolescents while higher doses had negative outcomes. In adult rats, nortriptyline did not significantly impact behavioral and neurochemical responses.

Summary: This study demonstrated that intravenous ketamine has significant short-term efficacy in reducing depressive symptoms in adolescents with depression, as well as being well tolerated acutely. Compared to an active placebo, ketamine resulted in a significant reduction in depressive symptoms within 24 hours of treatment.

Evidence for Use of Cannabinoids in Mood Disorders, Anxiety Disorders, and PTSD: A Systematic Review

Corneliu N. Stanciu et al.

Summary: In controlled trials assessing the effects of CBD and THC on affective disorders, anxiety disorders, and PTSD, the results showed insufficient evidence for their efficacy in managing these disorders. Therefore, medical cannabis should not be recommended for patients with these disorders. Further research is needed to explore the safety and efficacy of using cannabinoids to manage psychiatric disorders.

Synthesizing the Evidence for Ketamine and Esketamine in Treatment-Resistant Depression: An International Expert Opinion on the Available Evidence and Implementation

Summary: Replicated international studies have highlighted the human and societal costs of major depressive disorder, with monoamine-based antidepressants proving effective but many patients failing to fully recover. Ketamine and esketamine offer novel pharmacological treatments for treatment-resistant depression, but questions remain about their safety and effectiveness. International experts provide guidance on the use of these agents, while discussing areas of consensus and future research directions.

The effectiveness, safety and tolerability of ketamine for depression in adolescents and older adults: A systematic review

Joshua D. Di Vincenzo et al.

Summary: This article summarizes the literature on the effectiveness, safety, and tolerability of ketamine treatment for depression in different age groups. While most studies focus on adult populations, there is less research on adolescents and older adults. Although ketamine shows promising rapid antidepressant effects, the limited quantity and generally low quality of available studies restricts statistical syntheses and conclusions.

Repeated Dosing of Ketamine in the Forced Swim Test: Are Multiple Shots Better Than One?

Ridge G. Weston et al.

Summary: The successful repurposing of ketamine as an antidepressant has significant implications for clinical psychopharmacology. Studies suggest that repeated dosing of ketamine in rodents may have prominent depression-related effects and be more effective than a single dose. Further investigation may help optimize the use of ketamine in humans with MDD.

The forced swim test: Historical, conceptual and methodological considerations and its relationship with individual behavioral traits

Summary: The forced swim test is a commonly used method for evaluating antidepressant treatments and depression-like states in rodents, but its theoretical and predictive validity have been questioned. The test mainly assesses coping strategies in an inescapable situation, and proper interpretation of the behavior depends on its relationship with other behavioral traits.

Antidepressant-like effects of cannabidiol in a rat model of early-life stress with or without adolescent cocaine exposure

Cristian Bis-Humbert et al.

Summary: The study demonstrated that cannabidiol had antidepressant and anxiolytic effects in rats exposed to early-life stress, but had limited effects on cocaine. Cannabidiol played a positive role in reducing cocaine intake and reinstatement, but did not modulate CB receptors or BDNF proteins in the hippocampus.

Adolescent cocaine induced persistent negative affect in female rats exposed to early-life stress

Cristian Bis-Humbert et al.

Summary: The study found that adolescent cocaine exposure induced persistent negative affect in female rats during adulthood, particularly in those rats re-exposed to cocaine during abstinence. Maternal deprivation was identified as a suitable model for studying the impact of adolescent cocaine in female rats. This research highlights the risk of early drug initiation in adolescence for the emergence of negative reinforcement and vulnerability to cocaine addiction in female rats.

Increased negative affect when combining early-life maternal deprivation with adolescent, but not adult, cocaine exposure in male rats: regulation of hippocampal FADD

Cristian Bis-Humbert et al.

Summary: The study comparing the impact of maternal deprivation and cocaine exposure at different ages on affective behavior and hippocampal neuroplasticity regulation showed that adolescence is a more vulnerable period. This suggests that early accumulation of stress in life can anticipate negative behavioral outcomes associated with drug consumption.
