Summary: The study successfully obtained a high percentage of embryogenic calli from different genotypes of Miscanthus sinensis using mature seeds as explants, and developed a novel selective culture strategy to improve the efficiency of selecting resistant transformants. By optimizing the co-cultivation method and duration, the transformation frequency was significantly increased, providing an efficient tool for cultivar improvement and genetic analysis of Miscanthus sinensis or other Miscanthus species.
