A combined approach for single-cell mRNA and intracellular protein expression analysis

Summary: The SPARC method combines single-cell RNA sequencing with proximity extension assays to measure global mRNA and 89 intracellular proteins simultaneously in individual cells. Results show that mRNA expression does not accurately reflect protein abundance, with protein levels of transcription factors better predicting downstream effects. Overall, protein expression variation is lower than mRNA variation in single cells, but relative protein variation does not correlate with mRNA level.

The most important mechanism of heavy metal tolerance was found to be extracellular and intracellular sequestration, with an emphasis on the role of organic anions and cellular receptors. The functioning of chloroplasts, mitochondria, and the Golgi complex under heavy metal stress were also analyzed.

Summary: This study demonstrates that exogenous H2S significantly reduces arsenic accumulation in pea plants and promotes growth and photosynthetic parameters. H2S supply also reduces H2O2 and MDA levels in pea plants, as well as electrolyte leakage. Additionally, H2S enhances the activities of antioxidant enzymes and the AsA-GSH cycle enzymes, increasing levels of ascorbate, glutathione, and methylglyoxal.
