Summary: The study found that pharmacological inhibition of HO-1 activity in experimental tuberculosis can reduce bacterial loads, depending on the presence of T lymphocytes. Further research showed that the production of IFN gamma by T lymphocytes and the expression of NOS2 underlie this T-cell requirement for HO-1 inhibition, which enhances the IFN gamma-induced NOS2-dependent control of Mtb.

Drug repurposing for ligand-induced rearrangement of Sirt2 active site-based inhibitors via molecular modeling and quantum mechanics calculations

Summary: A computational screening of FDA-approved drugs identified 48 potential inhibitors of Sirt2, with Fluphenazine and Nintedanib showing substantial stability in Sirt2-drug complexes. The developed 3D-QSAR models supported the inhibitory potential of the drugs, highlighting Nintedanib as the most active compound with promising Sirt2 inhibition properties. Further in vitro investigation is required for drug development targeting Sirt2.

Adjunctive host-directed therapies for pulmonary tuberculosis: a prospective, open-label, phase 2, randomised controlled trial

Summary: This study conducted a phase 2 clinical trial in South Africa to evaluate the safety and preliminary efficacy of four host-directed therapies for tuberculosis. The results showed that CC-11050 and everolimus as adjunctive therapies have the potential benefits for tuberculosis patients, increasing the recovery of FEV1.

Doxycycline host-directed therapy in human pulmonary tuberculosis

Summary: The study demonstrated the pathological inhibition effect of doxycycline on matrix metalloproteinases (MMPs) in patients with pulmonary tuberculosis, and it could accelerate the restoration of gene expression. Doxycycline not only reduced the levels of MMPs in sputum of tuberculosis patients, but also inhibited the destruction of collagen and elastin, showing significant differences compared to placebo.
