Structure of pathological TDP-43 filaments from ALS with FTLD

Diana Arseni et al.

Summary: Abnormal aggregation of TDP-43 is a defining pathological hallmark in ALS and FTLD, as well as in other diseases. Cryo-electron microscopy revealed an amyloid-like filament structure comprising a single protofilament with a unique double-spiral-shaped fold in aggregated TDP-43, different from in vitro-formed filaments. The presence of glycine and neutral polar residues facilitates numerous turns and restricts beta-strand length, resulting in an absence of beta-sheet stacking and unique ligand-binding site possibilities. These findings enhance our understanding of ALS and FTLD molecular pathogenesis and inform the development of diagnostic and therapeutic agents targeting aggregated TDP-43.

Structural Identification of Individual Helical Amyloid Filaments by Integration of Cryo-Electron Microscopy-Derived Maps in Comparative Morphometric Atomic Force Microscopy Image Analysis

Liisa Lutter et al.

Summary: The presence of amyloid fibrils is a characteristic feature of various human disorders, and understanding the relationship between different structural polymorphs of amyloid fibrils and specific pathologies is a challenge. In this study, the researchers developed a method to identify individual fibril structures using atomic force microscopy (AFM) and high-resolution maps obtained from cryo-electron microscopy (cryo-EM). They successfully determined the structures of amyloid fibrils and found that certain fibrils were closely related to Alzheimer's disease. This study demonstrates the usefulness of AFM for individual particle structural analysis and provides insights into the structural polymorphism of amyloid fibrils.

Cryo-EM reveals structural breaks in a patient-derived amyloid fibril from systemic AL amyloidosis

Lynn Radamaker et al.

Summary: Systemic AL amyloidosis is a debilitating disease caused by the misfolding and fibrillation of immunoglobulin light chains (LCs). This study presents two ex vivo fibril structures of a lambda 3 LC derived from patient tissue, revealing the co-existence of two different fibril structures at different z-axial positions within the same fibril.

Summary: The study presents the cryo-EM structure of amyloid formed from the complete C-terminal TDP-43 low complexity domain and discusses the potential structural perturbations caused by phosphorylation and disease-related mutations. The findings reveal differences in the structural features of these fibrils compared to those generated from short protein fragments, providing insight into the mechanisms underlying neurodegenerative diseases like ALS and FTLD.

Cryo-EM structures of tau filaments from Alzheimer's disease with PET ligand APN-1607

Yang Shi et al.

Summary: A newly described PET ligand, APN-1607, was found to bind to specific sites in the Alzheimer's disease fold, potentially leading to the development of new ligands with increased specificity and binding activity. Additionally, tau filaments from posterior cortical atrophy (PCA) and primary age-related tauopathy (PART) were shown to be identical to those from AD.

The AD tau core spontaneously self-assembles and recruits full-length tau to filaments

Yari Carlomagno et al.

Summary: By identifying tau filament cores using cryoelectron microscopy, researchers found that the AD tau core has the intrinsic ability to spontaneously aggregate, drive aggregation of full-length tau, and template abnormal forms of tau. These findings provide a new approach to modeling tau aggregation and have significant implications for investigating tau transmission and developing biomarkers in vivo.

Co-factor-free aggregation of tau into seeding-competent RNA-sequestering amyloid fibrils

Pijush Chakraborty et al.

Summary: The study demonstrates that full-length tau protein can aggregate into seeding-competent amyloid fibrils without co-factors, which share similarities with tau fibrils isolated from the brains of patients with corticobasal degeneration. Additionally, the N-terminal 30 residues of tau are immobilized during fibril formation, in agreement with findings in pathological tau. Our findings suggest a critical advancement in understanding the molecular factors guiding aggregation towards disease-specific tau strains.
