A novel homozygous missense mutation of PMFBP1 causes acephalic spermatozoa syndrome

Mengmeng Lu et al.

Summary: In this study, a novel homozygous missense mutation of PMFBP1 was identified in an infertile male from a consanguineous family, leading to a significant decrease in expression of the mutant protein in sperm. This mutation may be a cause of acephalic spermatozoa syndrome, providing a basis for genetic counseling for the patient.

The missing linker between SUN5 and PMFBP1 in sperm head-tail coupling apparatus

Ying Zhang et al.

Summary: The study demonstrates the importance of CENTLEIN for the integrity and function of the head-to-tail coupling apparatus in sperm, with its absence leading to decapitated sperm and male sterility. It shows that CENTLEIN directly interacts with SUN5 and PMFBP1, playing a role in the assembly and integration of the head-to-tail coupling apparatus. Dysfunction of CENTLEIN may be associated with acephalic spermatozoa syndrome in humans.
