An open approach to systematically prioritize causal variants and genes at all published human GWAS trait-associated loci

Edward Mountjoy et al.

Summary: In this study, an open resource was introduced to systematically fine map and prioritize genes across published GWAS loci, aiming at identifying causal genes associated with complex traits. By training a machine learning model using genetics and functional genomics data, the authors were able to distinguish causal genes from neighboring genes, outperforming distance-based models. The prioritized genes were found to be enriched for known approved drug targets, suggesting their potential in drug development.

Rare genetic variants affecting urine metabolite levels link population variation to inborn errors of metabolism

Yurong Cheng et al.

Summary: This study explores the cumulative contribution of rare, exonic genetic variants on urine metabolite levels and identifies 30 genes related to inborn errors of metabolism, with shared expression in liver and kidney. By using in silico constraint-based modeling, the study correctly predicts the direction of metabolite changes, emphasizing the potential of linking population genetics to modeling.

Osteopontin in Cardiovascular Diseases

Kohsuke Shirakawa et al.

Summary: Advances in secondary prevention of cardiovascular diseases have greatly improved prognosis, but CVDs remain a leading cause of global mortality, warranting a re-evaluation of cardiovascular risk and optimal therapy. Inflammation, pro-thrombotic factors, and gene mutations have emerged as potential therapeutic targets for CVDs, with recent studies suggesting the possibility of anti-inflammatory therapy. Osteopontin (OPN) has shown dual roles in cardiovascular pathology, with acute increases having protective effects and chronic increases predicting a poor prognosis, highlighting its potential as a therapeutic target.
