Glucose starvation induces a switch in the histone acetylome for activation of gluconeogenic and fat metabolism genes

Wen-Chuan Hsieh et al.

Summary: Acetyl-CoA is a key intermediate that plays a critical role in coordinating gene expression with metabolic state. High levels of acetyl-CoA activate genes involved in cell growth or tumorigenesis through histone acetylation, but the role of histone acetylation under low levels of acetyl-CoA is not well understood. In a yeast starvation model, we observed a significant shift in the global occupancy of histone acetylation following carbon starvation. Histone acetylation marks relocate from growth-promoting genes to genes involved in gluconeogenesis and fat metabolism. This reallocation is mediated by the histone deacetylase Rpd3p and the acetyltransferase Gcn5p.

Glycerol-3-phosphate biosynthesis regenerates cytosolic NAD+ to alleviate mitochondrial disease

Shanshan Liu et al.

Summary: Through experimental validation, Gro3P synthesis has been identified as an endogenous pathway for NAD(+) regeneration, effectively restoring the effects of inhibited electron transport chain on cells and maintaining a certain consistency in different organisms. This discovery has the potential to become a therapeutic target for mitochondrial complex I diseases.

Reprogrammed lipid metabolism protects inner nuclear membrane against unsaturated fat

Anete Romanauska et al.

Summary: The study demonstrates that increased lipid unsaturation leads to a reprogramming of lipid storage metabolism across the nuclear envelope. Cells induce lipid droplet formation specifically from the distant membrane system, while suppressing lipid droplet formation at the inner nuclear membrane to avoid excessive lipid unsaturation.

Shortening of membrane lipid acyl chains compensates for phosphatidylcholine deficiency in choline-auxotroph yeast

Xue Bao et al.

Summary: Phosphatidylcholine (PC) is an essential membrane lipid component in most eukaryotes, but its necessity can be suppressed in yeast through cellular evolvability mechanisms involving chromosomal monosomy or a point mutation in the ACC1 gene. These adaptations lead to rewiring of lipid biosynthesis pathways and decreased activity of Acc1, resulting in altered acyl chain length and regulation of membrane fluidity.

MetaboAnalyst 5.0: narrowing the gap between raw spectra and functional insights

Zhiqiang Pang et al.

Summary: MetaboAnalyst version 5.0 has been introduced to facilitate comprehensive metabolomics data analysis and interpretation, with the aim of narrowing the gap from raw data to functional insights. Three new modules have been developed to assist in this goal, along with various other new functions and improvements in interface, graphics, and codebase. Users can now easily switch between compatible modules for a streamlined data analysis experience.
