Real world effects of COPD medications: a cohort study with validation against results from randomised controlled trials

Kevin Wing et al.

Summary: This study explored the application of non-interventional methods to real-world data for groups excluded from randomized controlled trials and found that the results were consistent with the TORCH trial for active comparator analyses. However, the study was unable to reproduce placebo-controlled results, underscoring the need for further research in this area.

Emulating Randomized Clinical Trials With Nonrandomized Real-World Evidence Studies First Results From the RCT DUPLICATE Initiative

Jessica M. Franklin et al.

Summary: The study found differences between RCT and corresponding RWE study populations, but showed that selecting active comparator therapies with similar indications and use patterns can enhance the validity of RWE. Despite efforts to closely emulate RCT design, agreement between RCT and RWE findings varied depending on the agreement metric used, indicating the need for further research to understand the frequency and contexts in which RWE findings match RCTs.

Real-world effects of medications for stroke prevention in atrial fibrillation: protocol for a UK population-based non-interventional cohort study with validation against randomised trial results

Emma Maud Powell et al.

Summary: This study aims to validate non-interventional methods for assessing the effectiveness of anticoagulant treatment by comparing the outcomes in an EHR trial-analogous cohort with the results of the ARISTOTLE trial. The study has been approved by the Independent Scientific Advisory Committee of the UK Medicines and Healthcare Products Regulatory Agency, and the results will be disseminated through scientific publications and relevant conferences.

Comparing Effect Estimates in Randomized Trials and Observational Studies From the Same Population: An Application to Percutaneous Coronary Intervention

Anthony A. Matthews et al.

Summary: By emulating a target trial with real-world data, this study found that observational data and trial results were largely consistent in the impact of bivalirudin versus heparin on death and myocardial infarction risk. However, limitations were identified in capturing less severe bleeding cases and estimating short-term mortality risk accurately.
