Evaluation of multiple analytical methods for SARS-CoV-2 surveillance in wastewater samples

Devrim Kaya et al.

Summary: This study evaluated 14 virus concentration protocols for quantification of SARS-CoV-2 in wastewater samples, finding the most effective combination for virus recovery. The impact of prolonged storage on enveloped virus decay was also assessed, along with the quantification of SARS-CoV-2 RNA in influent wastewater samples from wastewater treatment plants. The study may enhance understanding of the effective concentration and extraction of SARS-CoV-2 from wastewater.

Comparing analytical methods to detect SARS-CoV-2 in wastewater

Alba Perez-Cataluna et al.

Summary: Wastewater based epidemiology is a reliable strategy for assessing the COVID-19 pandemic, with recent studies showing the feasibility of detecting SARS-CoV-2 in wastewater. Different methods for concentration and extraction were evaluated, with the aluminum precipitation method coupled with automated nucleic extraction proving to be a sensitive and acceptable method for surveillance of SARS-CoV-2 in wastewater.

Evaluation of two rapid ultrafiltration-based methods for SARS-CoV-2 concentration from wastewater

E. Fores et al.

Summary: Quantitative measurements of SARS-CoV-2 in raw wastewater have been widely conducted since the pandemic, with efforts to evaluate different viral concentration methods to address supplier shortages. The study found significant differences in efficiency between two ultrafiltration-based methods for seeded viruses, while naturally occurring viruses showed no significant differences, suggesting the importance of testing both to evaluate concentration methods. The CP-Select (TM) method provided the highest concentration factor and proved to be fast, automatic, and suitable for working under BSL-2 measures.

Detection of SARS-CoV-2 in wastewater is influenced by sampling time, concentration method, and target analyzed

Nancy J. Pino et al.

Summary: Detection of SARS-CoV in wastewater is proposed for monitoring COVID-19 at the community level, but appropriate analytical methodologies have not been established. Different concentration methods showed varying efficiency in virus recovery, with ultrafiltration and aluminum precipitation being the most effective. The E gene was consistently detected in all samples analyzed, highlighting the need for clear protocols in wastewater surveillance.

Evaluating recovery, cost, and throughput of different concentration methods for SARS-CoV-2 wastewater-based epidemiology

Zachary W. LaTurner et al.

Summary: As the COVID-19 pandemic continues to impact communities worldwide, wastewater monitoring of the SARS-CoV-2 virus has emerged as a vital tool for predicting outbreaks. This study evaluated five different concentration methods for quantifying SARS-CoV-2 virus RNA in wastewater samples, with HA filtration and bead beating showing the best performance in terms of sensitivity and cost, providing guidance for labs conducting wastewater monitoring of SARS-CoV-2.
