An introduction to machine learning and analysis of its use in rheumatic diseases

Kathryn M. Kingsmore et al.

Summary: Machine learning is increasingly used in biomedicine, particularly in rheumatology, for classifying patients with RAIDs. While ML can identify relationships in data that were previously unrecognized, limitations such as sample size, accuracy of labelling, and lack of external validation datasets exist.

Predicting Response to Tocilizumab Monotherapy in Rheumatoid Arthritis: A Real-world Data Analysis Using Machine Learning

Fredrik D. Johansson et al.

Summary: The study developed a remission prediction model for TCZ monotherapy using machine learning algorithms and demonstrated good performance in real-world data. The conclusion showed that the remission prediction scores derived in RCTs discriminated patients in RWD as well as in RCTs, with further improvement in discrimination by retraining models in RWD.

Integrative Clinical, Molecular, and Computational Analysis Identify Novel Biomarkers and Differential Profiles of Anti-TNF Response in Rheumatoid Arthritis

Maria Luque-Tevar et al.

Summary: This study found that RA patients form distinct molecular clusters after receiving anti-TNF therapy, directly linked to their baseline clinical status. The clinical effectiveness of anti-TNF therapy varies among these molecular clusters, correlated with specific modulation of inflammatory response, restoration of altered oxidative status, reduction of NETosis, and reversal of related miRNAs. Integrated analysis of clinical and molecular profiles using machine learning can identify novel signatures as potential predictors of therapeutic response to TNFi therapy.

Predictive analytics for step-up therapy: Supervised or semi-supervised learning?

Mohammad Amin Morid et al.

Summary: The study compared supervised learning and semi-supervised learning in predicting whether rheumatoid arthritis patients would transition from first-line therapy to second-line therapy within a year. The results showed that the semi-supervised approach performed better in prediction, with higher precision and recall compared to the supervised approach.

Summary: This study evaluated the positive rate and prognostic significance of SMI in RA patients in remission with normal CRP and ESR levels. The results showed that the wrist joints had the highest SMI signal-positive rate, but the accuracy for predicting relapse was greatest for the MTP joints.

Advanced machine learning for predicting individual risk of flares in rheumatoid arthritis patients tapering biologic drugs

Asmir Vodencarevic et al.

Summary: A model was established using machine learning methods to estimate the probability of flares in rheumatoid arthritis (RA) patients tapering biological disease-modifying anti-rheumatic drugs (bDMARDs). By implementing a model selection approach based on nested cross-validation and stacking different classifiers, a powerful prediction model was created with an AUROC of 0.81. Key predictors of a flare included the percent dose change of bDMARDs, clinical disease activity, disease duration, and inflammatory markers.
