Therapy-induced polyploidization and senescence: Coincidence or interconnection?

Summary: Polyploid somatic cells play important roles in normal development and stress response. However, the transient polyploidy observed in early stages of tumors and during cancer treatment seems to be associated with tumor development and relapse. The mechanistic connection between therapy-induced senescence and polyploid formation needs to be urgently addressed, along with the question of reversibility of cancer cell senescence.

Summary: Recent data suggests that genotoxic agents in cancer therapy can cause genome shock and increase in cell size. This leads to genome duplication, formation of polyploid giant cancer cells, activation of an early embryonic program, and dedifferentiation of somatic cells. The giant cell life cycle, a proposed mechanism for malignant transformation, allows cells to restructure the genome and develop into blastocyst-like structures. These structures can generate resistant or metastatic daughter cells, and also enter embryonic diapause to survive environmental stress. Therapeutic agents can activate this developmental program, leading to recurrence or metastasis. Understanding the mechanisms of the giant cell life cycle offers new possibilities for therapeutic intervention.
