Humanization of a strategic CD3 epitope enables evaluation of clinical T-cell engagers in a fully immunocompetent in vivo model

A humanized CD3ε-knock-in mouse model for pre-clinical testing of anti-human CD3 therapy

Summary: The study introduces a murine genetic knock-in model expressing both murine and humanized CD3 epsilon-exon, which is responsive to manipulation with anti-human CD3 therapy, showing normal development and immune functionality. Results indicate no gross abnormalities in thymocyte development or T cell peripheral homeostasis, with comparable immune responses to wild type mice. Additionally, a graft-vs-host disease model driven by effector T cell responses revealed a wasting disease upon transfer of huCD3 epsilon(HET) T cells, demonstrating the effectiveness of this humanized CD3 murine model in pre-clinical tests of anti-human CD3 antibodies.

Immunotherapy combinations overcome resistance to bispecific T cell engager treatment in T cell-cold solid tumors

Summary: Our study in immunocompetent mouse models revealed that pretreatment tumor-associated T cell density is a critical factor in determining response to BiTE therapy. CD8+ T cells were identified as important targets and mediators of BiTE activity, while CD4(+) T cells played an antagonistic role in BiTE efficacy. In addition, therapeutic combinations such as ICB and 4-1BB agonism were found to synergize with BiTE treatment in poorly T cell-infiltrated, immunotherapy-refractory tumors.
