Clostridioides difficile toxins: mechanisms of action and antitoxin therapeutics

Shannon L. Kordus et al.

Summary: Clostridioides difficile produces up to three different toxins, which play a key role in the pathogenesis of colon infection. In this Review, Kordus, Thomas, and Lacy discuss the structure and function of these toxins and how this information guides new therapeutic approaches. Understanding the mechanisms of host-toxin interactions provides a foundation for developing novel strategies for the treatment and prevention of C. difficile infection.

Structural basis for selective modification of Rho and Ras GTPases by Clostridioides difficile toxin B

Zheng Liu et al.

Summary: Toxin B (TcdB) is a key factor in Clostridioides difficile infection (CDI), acting by glucosylating small GTPases in the Rho/Ras families. The recognition mechanism of TcdB towards Rho and R-Ras has been structurally elucidated, with adaptive residue changes in the glucosyltransferase domain (GTD) determining substrate preferences. Mutations disrupting GTPases binding reduce TcdB activity and toxicity, providing insights for therapeutic interventions for CDI.

Structural basis for CSPG4 as a receptor for TcdB and a therapeutic target in Clostridioides difficile infection

Peng Chen et al.

Summary: Chondroitin sulfate proteoglycan 4 (CSPG4) is identified as a potential receptor for C. difficile toxin B (TcdB) during C. difficile infections (CDIs). The cryo-EM structure of a TcdB-CSPG4 complex and CDI mouse models offer insights into the role of CSPG4 in CDIs and suggest a therapeutic strategy targeting TcdB.
