AAV capsid variants with brain-wide transgene expression and decreased liver targeting after intravenous delivery in mouse and marmoset

David Goertsen et al.

Summary: The study developed AAV capsids that achieved robust transgene expression in the brain with decreased liver targeting after non-invasive administration, enabling more targeted systemic gene delivery. This organ-specific targeting extends to marmosets, allowing for non-invasive gene delivery with distinct transgene expression patterns. The ability to cross the blood-brain barrier with neuronal specificity in rodents and non-human primates opens up new avenues for basic research and therapeutic possibilities beyond naturally occurring serotypes.

Reconstruction of the Hypothalamo-Neurohypophysial System and Functional Dissection of Magnocellular Oxytocin Neurons in the Brain

Bin Zhang et al.

Summary: The study reveals the three-dimensional architecture of the hypothalamo-neurohypophysial system (HNS) using viral tracing and whole-brain imaging, uncovering complexity of MNCs collateral projections within the brain. It also demonstrates that subsets of MNCs collaterally project to multiple extrahypothalamic regions, providing structural and functional evidence for coordinating both peripheral and central oxytocin-mediated actions.

Neural control of affiliative touch in prosocial interaction

Ye Emily Wu et al.

Summary: The ability to help and care for others is crucial for social cohesiveness and the well-being of social species, including humans. Affiliative social touch, such as allogrooming, is a major prosocial behavior that provides comfort to others and strengthens social bonds. This study demonstrates that mice exhibit prosocial comforting behavior and reveals a neural circuit mechanism that underlies the encoding and control of affiliative touch during social interactions.

Enhancer viruses for combinatorial cell-subclass-specific labeling

Lucas T. Graybuck et al.

Summary: New genomic single-cell analysis methods have enabled rapid cell type identification, but efficient experimental access to these cell types remains a challenge. By collecting chromatin accessibility data and identifying enhancers specific to cell subclasses and types, researchers have been able to label different neuronal subclasses in the mouse cortex using recombinant adeno-associated viruses (AAVs). Through coupling enhancer viruses expressing recombinases with a newly generated transgenic mouse, strong labeling of multiple neuronal classes/subclasses can be achieved in the brain of a single transgenic animal, providing unprecedented flexibility and specificity for investigating cell types in the mouse brain and beyond.
