The emerging roles of circRNAs in cancer and oncology

Lasse S. Kristensen et al.

Summary: Circular RNAs (circRNAs) are a novel class of primarily non-coding RNAs with increasingly recognized roles in cancer development and progression through diverse mechanisms of action. In this review, the authors summarize the current understanding of circRNA biogenesis, regulation, physiological functions and pathophysiological roles in cancer. They also discuss the clinical potential of circRNAs as biomarkers, therapeutic agents and drug targets in oncology, as well as the research controversies, technical issues, and biological knowledge gaps that need to be addressed.

Circ_0003972 Promotes the Proliferation and Inflammation of Fibroblast-like Synovial Cells in Rheumatoid Arthritis through Regulation of the miR-654-5p/FZD4 Axis

Juan Li et al.

Summary: The study revealed that circ_0003972 was highly expressed in RA patients and HFLS-RA cells, and its knockdown inhibited proliferation and inflammation while promoting apoptosis in TNF alpha-induced HFLS-RA cells. MiR-654-5p was downregulated in RA patients and HFLS-RA cells, and could be sponged by circ_0003972. Furthermore, FZD4, targeted by miR-654-5p and positively regulated by circ_0003972, reversed the effects of miR-654-5p on proliferation, inflammation, and apoptosis in TNF alpha-induced HFLS-RA cells.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Circular RNAs: Promising biomarkers for cancer diagnosis and prognosis

Guohao Wei et al.

Summary: circRNAs, characterized by covalent bonds linking 3' and 5' ends, act as miRNA sponges in tumorigenesis. With their stable, conserved and cell/tissue-specific characteristics, circRNAs have shown potential as molecular markers for cancer, allowing for convenient and noninvasive diagnostic methods. Despite unclear mechanisms, circRNAs play a universal and critical role in regulating cancer cellular processes, suggesting their potential as biomarkers for cancer diagnosis, treatment, and prognosis.

MAPK4 promotes prostate cancer by concerted activation of androgen receptor and AKT

Tao Shen et al.

Summary: The study revealed that MAPK4 promotes prostate cancer growth and castration resistance by activating GATA2/AR and AKT pathways. Activation of both GATA2/AR and AKT is necessary for MAPK4 tumor-promoting activity.

RNF168 regulates R-loop resolution and genomic stability in BRCA1/2-deficient tumors

Parasvi S. Patel et al.

Summary: Germline mutations in BRCA1 and BRCA2 genes increase the risk of breast and ovarian cancer, while the loss of RNF168 can protect Brca1-mutant mice against mammary tumorigenesis. Studies have shown that RNF168 deficiency leads to accumulation of R-loops in BRCA1/2-mutant breast and ovarian cancer cells, causing cell death.
