Biology and evolution of bacterial toxin-antitoxin systems

Dukas Jurenas et al.

Summary: Toxin-antitoxin systems, composed of toxins and antitoxins, are widely found in bacterial genomes. Recent studies have challenged the traditional understanding of these systems and focused more on their role in host-genetic element interactions.

Structure-based design of peptides that triggerStreptococcus pneumoniaecell death

Sung-Min Kang et al.

Summary: This study reveals a novel antimicrobial agent that activates HigB toxin to degrade mRNA as an antibacterial strategy against antibiotic-resistant Streptococcus pneumoniae, with a mechanism different from current antibiotics. Additionally, the structure of the HigBA complex and potential catalytic residues of HigB were identified, providing insights into the transcriptional regulation mechanisms and allosteric inhibition of HigB activity.

ClpXP-mediated Degradation of the TAC Antitoxin is Neutralized by the SecB-like Chaperone in Mycobacterium tuberculosis

Pauline Texier et al.

Summary: Bacterial toxin-antitoxin systems, including TAC modules with SecB-like chaperones, are widespread in bacterial genomes. Deletion of endogenous AAA(+) proteases bypasses the need for a dedicated chaperone, and the ClpXP1 P2 complex is identified as the main protease involved in TAC antitoxin degradation.

Type VII Toxin/Antitoxin Classification System for Antitoxins that Enzymatically Neutralize Toxins

Summary: This study identified three new TA systems where the antitoxin is an enzyme that chemically modifies the toxin to neutralize its activity. The suggestion to introduce the type VII TA system will help classify enzyme-antitoxin reaction-based TA systems.

HigB1 Toxin in Mycobacterium tuberculosis Is Upregulated During Stress and Required to Establish Infection in Guinea Pigs

Arun Sharma et al.

Summary: The expansion of Toxin Antitoxin modules in Mycobacterium tuberculosis has been extensively studied, highlighting their importance in stress response and pathogenesis. Specifically, the HigB1 toxin plays a crucial role in stress adaptation and pathogenesis of M. tuberculosis, as well as in regulating the expression of virulence-related genes. Deletion of higB1 results in reduced bacterial loads and pathological damage in infected guinea pigs, emphasizing the significance of the higBA1 locus during infection in host tissues.

Shutoff of host transcription triggers a toxin-antitoxin system to cleave phage RNA and abort infection

Chantal K. Guegler et al.

Summary: Research has found that toxin-antitoxin system toxIN can protect E. coli against bacteriophages, with ToxN being activated following T4 infection to inhibit phage development by cleaving viral mRNAs. Transcriptional shutoff is crucial for ToxN activation, but toxIN does not provide strong protection against T7 phage, indicating a critical trade-off in blocking host transcription.
