Reverse β-oxidation pathways for efficient chemical production

Katia Tarasava et al.

Summary: Microbial production of fuels, chemicals, and materials is important for reducing greenhouse gas emissions and promoting a sustainable bioeconomy. Synthetic biology allows modification of metabolic pathways for desired product synthesis, but current pathways have limitations and are regulated by complex mechanisms. The reverse beta-oxidation pathway offers an alternative solution with high efficiency and versatility in producing bioproducts, and its application can be extended to different organisms.

Hexanol biosynthesis from syngas by Clostridium carboxidivorans P7 - product toxicity, temperature dependence and in situ extraction

Patrick Kottenhahn et al.

Summary: Clostridium carboxidivorans converts syngas into industrial alcohols like hexanol, but product toxicity may limit titers. Adding oleyl alcohol as an extraction solvent increases hexanol production, with the highest efficiency achieved compared to other solvents. Higher hexanol tolerance is observed at lower temperatures, while cell membranes adapt to higher temperature by incorporating more saturated fatty acids, with no significant effect on hexanol production. Corn oil and sunflower seed oil are tested as alternative extraction solvents, showing similar hexanol titers as oleyl alcohol.

Toward sustainable, cell-free biomanufacturing

Blake J. Rasor et al.

Summary: Industrial biotechnology is an attractive approach for producing low-cost fuels and products from sustainable resources, but cells pose limitations on product synthesis and release. Cell-free biotechnology, utilizing biological machinery harvested from cells, offers advantages such as shorter development times, higher production rates, and tolerance to toxic molecules. Expanding the practice of cell-free biomanufacturing could lead to innovative approaches for producing green chemicals on an industrial scale.

An orthogonal metabolic framework for one-carbon utilization

Alexander Chou et al.

Summary: The study introduces an alternative approach using synthetic pathways for C1 bioconversion, generating multicarbon products directly from formate, formaldehyde and methanol while being orthogonal to the host metabolic network. Variants of the FORCE pathways were evaluated through thermodynamic and stoichiometric analyses, showing promising results both in vitro and in vivo. This synthetic metabolic pathway demonstrates the potential for C1 compound utilization and integration with host metabolism for synthetic methylotrophy.

An integrated in vivo/in vitro framework to enhance cell-free biosynthesis with metabolically rewired yeast extracts

Blake J. Rasor et al.

Summary: Cell-free systems using genetically rewired cell extracts enable faster and more efficient design of biosynthetic pathways for sustainable chemical synthesis. This integrated framework allows for significant increases in product titers and volumetric productivities through systematic optimization of the cell-free reaction environment. The approach is generalizable to other biosynthesis pathways, showcasing the potential for synthetic biology prototyping and cell-free biomanufacturing.
