New derivatives of the iridoid specioside from fungal biotransformation

Nadla Soares Cassemiro et al.

Summary: Fungus-mediated biotransformation of the iridoid specioside was conducted using seven fungi, resulting in a total of nineteen different analogs. Main metabolite observed was the non-glycosylated specioside, with other analogs produced from ester hydrolysis, hydroxylation, methylation, and hydrogenation reactions. All fungi-mediated biotransformation yielded non-glycosylated specioside and coumaric acid, showing the ability of the fungi to perform hydroxylation, glycosidic, and ester hydrolysis reactions from glycosylated iridoid.

De novo biosynthesis of bioactive isoflavonoids by engineered yeast cell factories

Quanli Liu et al.

Summary: Isoflavonoids are industrially important plant natural products, but their low abundance and structural complexity limit their availability. By engineering Saccharomyces cerevisiae metabolism to efficiently produce daidzein, a core chemical scaffold for isoflavonoid biosynthesis, researchers demonstrate its application for producing bioactive glucosides from glucose.

Catalytic flexibility of rice glycosyltransferase OsUGT91C1 for the production of palatable steviol glycosides

Jinzhu Zhang et al.

Summary: This study characterizes a rice enzyme OsUGT91C1 that efficiently catalyzes beta (1-2) glucosylation, and engineers it to enhance the production of naturally rare but palatable steviol glycosides Reb D and Reb M. Through structural insights, the enzyme's flexibility in binding steviol glycoside substrates allows for targeted modifications, making it a promising catalyst for industrial-scale production.

Metabolic network remodelling enhances yeast's fitness on xylose using aerobic glycolysis

Xiaowei Li et al.

Summary: In this study, a combination of metabolic engineering, systems biology, and adaptive laboratory evolution was used to gain insights into how yeast rewires cellular processes to efficiently adapt to metabolic transitions from glucose to xylose. Through metabolic engineering, enhanced cell growth on xylose was achieved, with transcriptome analysis revealing involvement of multiple pathways in cellular reprogramming. Genome resequencing and reverse engineering identified key factors like SWI/SNF chromatin remodeling, osmotic response, and aldehyde reductase in improving growth on xylose, with glycerol-3-phosphate and xylitol identified as key metabolites affecting cellular adaptation.
