The calcium dynamics of human dental pulp stem cells stimulated with tricalcium silicate-based cements determine their differentiation and mineralization outcome

Summary: This study investigated the Ca2+ responses of human dental pulp stem cells (hDPSCs) stimulated with two different tricalcium silicate-based (TCS-based) dental biomaterials, Biodentine and ProRoot white MTA. The findings showed that Biodentine released more Ca2+ in the culture medium compared to ProRoot WMTA, leading to altered intracellular Ca2+ dynamics and enhanced cellular mineralization potential. The biomaterial-specific Ca2+ dynamics influenced stem cell-related gene expression, cellular differentiation, and mineralization outcomes.

Summary: The design of bioink must consider viscoelastic properties, rheological properties, injection speed, and printing nozzle diameter, as these are related to cell behavior and survival rate during and after printing. Mechanosensing plays a strong role in influencing epigenetics to modify final cell phenotype, which can impact replacement tissue. Biotechnologists in tissue engineering must take into account the biophysical properties and biological conditions of materials used, as well as material delivery and maturation modes, to ensure appropriate materials mimicking native tissue development.

Dental stem cell signaling pathway activation in response to hydraulic calcium silicate-based endodontic cements: A systematic review of in vitro studies

Summary: In vitro studies have shown that hydraulic calcium silicate-based cements elicit a favorable biological response from a variety of dental stem cells, supporting their use in biologically-based endodontic procedures. Signaling pathways such as MAPK, NF-kappa B, Wnt/β-catenin, BMP/Smad, and CAMKII have been proposed as potential mediators in the biological interaction between dental stem cells and hydraulic calcium silicate-based cements. Understanding these signaling processes could help in developing new biomaterial compositions targeted at enhancing tissue repair mechanisms through biologically-based procedures.
