Parvovirus-Based Combinatorial Immunotherapy: A Reinforced Therapeutic Strategy against Poor-Prognosis Solid Cancers

Summary: Oncolytic virotherapy using viruses with cancer-destroying capacities has shown promising results in both preclinical models and clinical studies, particularly the rat H-1 parvovirus. These viruses not only induce tumor cell death but also stimulate the immune system and reverse tumor-driven immune suppression, enhancing the efficacy of other cancer immunotherapies. Challenges and prospects of using parvoviruses in immunotherapy against therapy-refractory solid malignancies are discussed, highlighting the potential to modulate the tumor microenvironment and promote antitumor immune responses.

Summary: In-depth understanding of the life cycle of H-1 parvovirus (H-1PV) is crucial for the rational design of combination strategies and development of more effective viruses. The identification of laminin gamma 1 (LAMC1) as a crucial modulator of H-1PV infection highlights the importance of laminin family members in cell attachment and entry. Elevated levels of LAMC1 are found to correlate with increased H-1PV oncolytic activity in various cancer cell lines.
