Akt1-associated actomyosin remodelling is required for nuclear lamina dispersal and nuclear shrinkage in epidermal terminal differentiation

Clare Rogerson et al.

Summary: The study shows that the dispersal of phosphorylated Lamin A/C in differentiating keratinocytes is Akt1-dependent and involves the cytoskeletal proteins actin, Arp3, and Myh9. Disruption of actin filament polymerisation, nucleation, or myosin IIa activity prevents cytoplasmic Lamin A/C structure formation. Live imaging reveals a rapid nuclear volume reduction step before final destruction, dependent on Akt1 and cytoskeleton-associated proteins.

NIX initiates mitochondrial fragmentation via DRP1 to drive epidermal differentiation

Cory L. Simpson et al.

Summary: This study reveals that regulated breakdown of mitochondria is critical for epidermal development, with keratinocytes in the upper layers initiating mitochondrial fragmentation, depolarization, and acidification. The mitophagy receptor NIX and DRP1 play essential roles in driving epidermal morphogenesis by promoting mitochondrial degradation in the terminal differentiation of keratinocytes.

Summary: This study used intravital imaging and intracellular probes to reveal the crucial processes of SG1 cell death, showing that intracellular Ca2* elevation and acidification induction are essential for corneocyte formation, regulated by the transient receptor potential cation channel V3.
