Reduced innate lymphoid cells in the endometrium of women with endometriosis

Takuya Sugahara et al.

Summary: The study found that the proportion of ILC2 and 3 was reduced in the endometrium of patients with endometriosis, while ILCs were increased in ovarian endometrioma. This may indicate a new immunological approach to understand the pathophysiology of endometriosis.

IL-33 and Soluble ST2 Are Associated With Recurrent Spontaneous Abortion in Early Pregnancy

Long Zhao et al.

Summary: This study investigated the abnormal expression of cytokines in the process of inducing Recurrent spontaneous abortion (RSA). The results showed that the serum concentrations of IL-33 and ST2 were significantly higher in the RSA group compared to the healthy control group. Additionally, the serum concentrations of IL-33 and ST2 were not significantly correlated with the number of weeks of pregnancy.

Targeting androgen signaling in ILC2s protects from IL-33-driven lung inflammation, independently of KLRG1

Eve Blanquart et al.

Summary: This study investigates the impact of androgens on the development and function of innate lymphoid cells in female lungs. The findings suggest that androgens negatively control innate lymphoid cell homeostasis and inflammation in a cell-intrinsic manner. The inhibitory receptor killer cell lectin-like receptor G1 is found to play a dispensable role in androgen-mediated inhibition of innate lymphoid cells.

Summary: Interleukin-33 (IL-33) is an immune signaling molecule that plays a role in various diseases. It activates different cells to initiate and amplify immune responses and inflammation. IL-33 has multiple regulatory mechanisms to control its activity and function.

Targeting the Epithelium-Derived Innate Cytokines: From Bench to Bedside

Jongho Ham et al.

Summary: Epithelial cells produce alarmin cytokines when exposed to external stimuli, which alert the immune system and mobilize immune defense mechanisms. Although primarily studied in allergic and infectious diseases, recent research suggests that these cytokines also play important roles in other pathological conditions.

Increased levels of serum IL-33 is associated with adverse maternal outcomes in placenta previa accreta

Sibel Ozler et al.

Summary: The study concluded that increased levels of maternal serum IL-33 and history of previous cesarean section were significantly associated with placenta previa accreta (PPA). Furthermore, elevated maternal serum IL-33 levels were linked to cesarean hysterectomy and massive blood transfusion in PPA cases, suggesting a potential role of IL-33 in abnormal placental invasion.

Estrogen receptor-α signaling increases allergen-induced IL-33 release and airway inflammation

Jacqueline-Yvonne Cephus et al.

Summary: In this study, estrogen receptor-alpha signaling was found to increase IL-33 release and ILC2-mediated airway inflammation. Esr1-deficient mice showed significantly reduced IL-5, IL-13 production, BAL eosinophils, and IL-33 release, while PPT and 17β-estradiol increased IL-33 gene expression and release.

Development of an ultra-sensitive human IL-33 biomarker assay for age-related macular degeneration and asthma drug development

Summary: Research has shown that most current commercial hIL-33 detection assays lack the sensitivity to accurately detect reduced hIL-33 at lower biological relevant levels (sub-to-low pg/mL), especially in the presence of human sST2 (hsST2). As a result, researchers developed and validated a highly sensitive and specific enzyme-linked immunosorbent assay (ELISA) capable of accurately quantifying both total and reduced hIL-33 levels even in the presence of high levels of sST2.

Asthma-associated genetic variants induce IL33 differential expression through an enhancer-blocking regulatory region

Ivy Aneas et al.

Summary: This study identified a 5 kb regulatory region within the IL33 locus that mediates allele-specific regulatory activity and IL33 expression, potentially contributing to genetic risk of asthma. Furthermore, the asthma-associated SNP rs1888909 was found to be associated with IL33 gene expression in human airway epithelial cells and IL-33 protein expression in human plasma, possibly through differential binding of OCT-1 to the asthma-risk allele.

Il4ra-independent vaginal eosinophil accumulation following helminth infection exacerbates epithelial ulcerative pathology of HSV-2 infection

Alisha Chetty et al.

Summary: Research has shown that acute helminth infection induces a type 2 immune profile in the female genital tract, leading to increased epithelial ulceration and pathology during subsequent herpes simplex virus infection. This is mediated by an inflammatory circuit involving eosinophils, IL-33, and IL-5, which enhances the risk of vaginal epithelial necrosis and pathology.

IL-33 activates group 2 innate lymphoid cell expansion and modulates endometriosis

Jessica E. Miller et al.

Summary: The study found that IL-33 contributes to the expansion of ILC2s in both humans and murine models, regulating the microenvironment of endometriosis lesions, and driving severe endometriosis symptoms such as inflammation, lesion proliferation, and fibrosis. IL-33 neutralization shows promise as a potential novel therapeutic avenue for treating the debilitating symptoms of endometriosis.

IL-33 causes thermogenic failure in aging by dysfunctional adipose ILC2

Emily L. Goldberg et al.

Summary: The study highlighted the decrease in type 2 innate lymphoid cells (ILC2) in aged adipose tissue, leading to changes in the immune system and thermogenic failure. Supplementation of IL-33 and transplantation of adult ILC2 cells can protect old mice from the effects of cold stress to some extent.
