Improving the freeze-drying survival rate of Lactobacillus plantarum LIP-1 by increasing biofilm formation based on adjusting the composition of buffer salts in medium

Summary: Adding different buffer salts in the culture medium had a significant impact on the freeze-drying survival rate of Lactobacillus plantarum LIP-1, possibly due to the promotion of biofilm formation. Potassium ions in buffer salt increased the expression of the luxS gene, leading to increased synthesis of quorum sensing signal AI-2 and upregulation of the cysE gene, which helps promote biofilm formation. Conversely, addition of the biofilm inhibitor D-galactose down-regulated luxS and cysE genes, reducing biofilm formation and freeze-drying survival rate. Promoting biofilm formation using appropriate buffer salts may lead to better freeze-drying survival rates.

The study suggests a close relationship between bacteriocin synthesis and the LuxS-mediated quorum sensing system in co-cultivation.

Summary: Understanding aquatic pathogens in sediments or aquacultural water is crucial for protecting public health from soilborne and waterborne diseases. Quorum sensing (QS) has been increasingly reported in biological wastewater treatment processes due to its roles in biofilm development and bacterial aggregation. The widely used QS signal AI-2 is primarily involved in interspecies communication. The RbsB protein of Streptococcus agalactiae may mediate quorum sensing signal uptake via interaction with AI-2, and further studies suggest that RbsB plays a role in the response of S. agalactiace to AI-2.
