Summary: The catalytic synthesis of P- and axially chiral biaryl phosphine oxides was achieved using Cp*Ir-catalyzed asymmetric C-H coupling, with the stereoselectivity controlled by chiral carboxylic amides. This review summarizes recent advances in transition-metal-catalyzed enantioselective desymmetric C-H functionalization reactions via coordination-assisted metal insertion, covering the literature from 2008 to June 2021. Mechanistic pathways and stereocontrol modes are discussed where appropriate.

Summary: The study presents ruthenium(II)-catalyzed enantioselective C-H functionalization reactions of sulfoximines with sulfoxonium ylides. The combination of [RuCl2(p-cymene)](2) and a pseudo-C-2-symmetric binaphthyl monocarboxylic acid yielded S-chiral products in 76:24 to 92:8 er.

Synthesis of Chiral Sulfoxides via Pd(II)-Catalyzed Enantioselective C-H Alkynylation/Kinetic Resolution of 2-(Arylsulfinyl)pyridines

Summary: This study reports a Pd(II)-catalyzed enantioselective C-H alkynylation of 2-(arylsulfinyl)pyridines via kinetic resolution using inexpensive and commercially available L-pGlu-OH as a chiral ligand. A wide range of 2-(arylsulfinyl)pyridines were compatible with this protocol, yielding alkynylation products with high yields and high enantioselectivities (up to 99% ee). Furthermore, the enantioenriched products can be easily transformed into various types of chiral sulfoxide scaffolds while retaining enantiopurity.

Desymmetric Enantioselective Reduction of Cyclic 1,3-Diketones Catalyzed by a Recyclable P-Chiral Phosphinamide Organocatalyst

Summary: The P-stereogenic phosphinamides have been demonstrated as powerful organocatalysts for the desymmetric enantioselective reduction of cyclic 1,3-diketones, providing a useful method for the synthesis of chiral cyclic 3-hydroxy ketones. The protocol shows a broad substrate scope and high enantioselectivities and diastereoselectivities for the chiral cyclic 3-hydroxy ketone products. Mechanistic studies revealed that an intermediate formed from P-stereogenic phosphinamide and catecholborane is the real catalytically active species, which paves the way for designing and developing other reactions using P-stereogenic phosphinamides as new organocatalysts.

Iridium-Catalyzed Enantioselective C-H Borylation of Diarylphosphinates

Efficient Synthesis of Sulfur-Stereogenic Sulfoximines via Ru(II)-Catalyzed Enantioselective C-H Functionalization Enabled by Chiral Carboxylic Acid

Summary: An undeveloped Ru(II)-catalyzed enantioselective C-H functionalization involving an enantiodetermining C-H cleavage step has been described in this study. Using a novel class of chiral binaphthyl monocarboxylic acids as chiral ligands, a broad range of sulfur-stereogenic sulfoximines were synthesized in high yields with excellent enantioselectivities (up to 99% yield and 99% ee) via desymmetrization, kinetic resolution, and parallel kinetic resolution. Furthermore, the resolution products can be easily transformed into chiral sulfoxides and key intermediates for kinase inhibitors.

Kinetic resolution of sulfur-stereogenic sulfoximines by Pd(ii)-MPAA catalyzed C-H arylation and olefination

Summary: A direct Pd(ii)-catalyzed kinetic resolution of heteroaryl-enabled sulfoximines through an ortho-C-H alkenylation/arylation of arenes has been developed, providing access to a wide range of enantiomerically enriched unreacted aryl-pyridyl-sulfoximine precursors and C(aryl)-H alkenylation/arylation products in good yields with high enantioselectivity. The coordination of the directing group, ligand effect, geometry constraints, and the transient six-membered concerted-metalation-deprotonation species dictate the stereoselectivity, which is validated by DFT studies.
