Engineering bionanoparticles for improved biosensing and bioimaging

Summary: Bionanoparticles, such as extracellular vesicles and protein nanoparticles, are ideal platforms for biosensing and bioimaging applications due to their high density surface functionalization and large loading capacity. By scaffolding large numbers of sensing and detection modules onto these nanoparticles, enhanced analyte affinity and specificity, as well as signal amplification for highly sensitive detection even at low analyte concentrations, can be achieved. Recent examples in literature demonstrate the potential of bionanoparticles for bioimaging and biosensing, highlighting their ability to generate highly sensitive detection devices with impressive signal amplification.

Summary: An amplified electrochemiluminescence ratiometric biosensor for p53 DNA sequence assay was developed using gold@carbon dots and gold@luminol nanoparticles, along with an enzyme-free DNA walker for signal amplification. The sensor exhibited high sensitivity and accuracy, with a lower limit of 0.34 fM for target detection. The use of Au@luminol and Au@CDs offered cost-effective, biocompatible, and easily labeled advantages, providing a new platform for ratiometric ECL biosensors.
