Targeted Solvatochromic Fluorescent Probes for Imaging Lipid Order in Organelles under Oxidative and Mechanical Stress

Dmytro Danylchuk et al.

Summary: A series of solvatochromic probes have been designed for specific targeting of different organelles in live cells, allowing the detection of variations in lipid order and quantitative description of lipid order in each organelle. These probes revealed the characteristics of organelles under stress conditions, with mitochondria and lysosomes particularly sensitive to stress. The new probes provide a powerful toolbox for monitoring cellular response to physical and chemical stimuli at the level of individual organelle membranes.

Immunometabolism of Macrophages in Bacterial Infections

Gael Galli et al.

Summary: Macrophages play crucial roles in tissue homeostasis, inflammation, and host defense by recognizing microbial or danger signals and eliciting immune responses. The interaction between cellular metabolism and macrophage innate immunity involves metabolic adaptations that modulate immune signaling and macrophage function. Intracellular bacterial pathogens can exploit macrophage metabolic pathways to evade immune defenses. This review highlights recent evidence on host-bacterial immunometabolic interactions.

Quantitative and Multiplexed Fluorescence Lifetime Imaging of Intercellular Tensile Forces

Puspam Keshri et al.

Summary: Fluorescence lifetime-based imaging and quantification of intercellular molecular tensions offer a novel approach to simultaneously measure tensile forces among multiple ligand-receptor pairs. This method was validated and DNA tension probes were developed for imaging E-cadherin-mediated tension, which were further applied to quantify the correlations between E-cadherin and N-cadherin tensions during an epithelial-mesenchymal transition process. The modular design of these probes has the potential to study the mechanical features of various physiological and pathological processes.
