The detyrosination/re-tyrosination cycle of tubulin and its role and dysfunction in neurons and cardiomyocytes

Chadni Sanyal et al.

Summary: Among various post-translational modifications of microtubules, the detyrosination/re-tyrosination cycle specifically occurs in tubulin. This cycle involves enzymatic removal and re-addition of a tyrosine residue at the C-terminus of alpha-tubulin, and also the conversion of detyrosinated tubulin to Delta 2-tubulin. The cycle regulates microtubule dynamics and cargo trafficking, and plays a crucial role in neuronal health and heart function. Dysregulation of the cycle is associated with neurodegeneration and heart failure in humans.

Pathogenic role of delta 2 tubulin in bortezomib-induced peripheral neuropathy

Maria Elena Pero et al.

Summary: The study demonstrates that bortezomib affects microtubule stability in sensory neurons by promoting the accumulation of delta 2 tubulin, leading to peripheral neuropathy. Furthermore, it suggests a previously unrecognized pathogenic role for D2 in bortezomib-induced peripheral neuropathy, potentially through altered regulation of mitochondria motility.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

MARK4 controls ischaemic heart failure through microtubule detyrosination

Xian Yu et al.

Summary: Myocardial infarction leads to compromised cardiac function and chronic heart failure, necessitating the development of new drugs to improve cardiomyocyte contractility. Recent research highlights the role of MARK4 in regulating microtubule detyrosination in cardiomyocytes, making it a promising therapeutic target for improving cardiac function post-myocardial infarction.
