Strategies for developing complex multi-component in vitro tumor models: Highlights in glioblastoma

Thomas J. DePalma et al.

Summary: In recent years, bioengineered in vitro models of cancer have been widely used for studying disease mechanisms, testing drugs, and developing personalized treatment options. This review discusses the considerations in developing accurate model systems that mimic the tumor microenvironment and can answer specific scientific questions. The importance of cell sourcing and biomaterial selection is emphasized, and examples of model form factors and biofabrication techniques are provided. The review also explores the application of these models in the context of glioblastoma, illustrating how they contribute to cancer biology research and drug development.

Engineered human blood-brain barrier microfluidic model for vascular permeability analyses

Cynthia Hajal et al.

Summary: This protocol describes an in vitro model of the human blood-brain barrier, self-assembled within microfluidic devices from stem-cell-derived or primary brain endothelial cells, and primary brain pericytes and astrocytes. The model features relevant cellular organization and morphological characteristics, and can be used to assess pathophysiological molecular transport mechanisms and design targeted therapies for neurological disorders.

Low nanogel stiffness favors nanogel transcytosis across an in vitro blood-brain barrier

Lais Ribovski et al.

Summary: This study investigated the effect of nanoparticle stiffness on transport across the blood-brain barrier, finding that more densely cross-linked nanogels showed higher uptake by brain endothelial cells, while less densely cross-linked nanogels demonstrated higher transcytotic potential. These findings suggest that nanogel stiffness has opposing effects on nanogel uptake and transcytosis at the BBB.

The cancer glycocalyx mediates intravascular adhesion and extravasation during metastatic dissemination

Giovanni S. Offeddu et al.

Summary: Offeddu et al. used a microfluidic vascular model to study the effects of selectively removing glycocalyx components from cancer cells. They found that cancer-associated hyaluronic acid and CD44 mediate firm adhesion to endothelial cells and extravasation during metastasis.

Meet me halfway: Are in vitro 3D cancer models on the way to replace in vivo models for nanomedicine development?

Sabina Pozzi et al.

Summary: Three-dimensional cancer models have been developed to study a wide range of biological applications, including cell signaling pathways, metastatic cascade steps, and personalized medicine, in vitro. By combining different cell types and external stimuli, these models can more accurately mimic the biology and pathophysiology of in vivo tumors.

Microheart: A microfluidic pump for functional vascular culture in microphysiological systems

Giovanni S. Offeddu et al.

Summary: Advances in microphysiological systems have led to a demand for long-term cell culture under physiological flow conditions, and a microfluidic-based pump has been presented for controlled media delivery in vascular microphysiological applications. The pump's performance was characterized in various microfluidic systems, and a theoretical framework along with a finite element model were developed to predict its performance in different fluidic configurations. The use of the pump for microvascular physiological studies demonstrated its utility in replicating vascular fluid transport phenomena in microphysiological systems.

Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Toshiro Hara et al.

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.

The CCL2-CCR2 astrocyte-cancer cell axis in tumor extravasation at the brain

Cynthia Hajal et al.

Summary: The study demonstrates that astrocytes promote cancer cell extravasation across the blood-brain barrier by secreting CCL2 and the CCL2-CCR2 astrocyte-cancer cell axis plays a fundamental role in this process.

Human physiomimetic model integrating microphysiological systems of the gut, liver, and brain for studies of neurodegenerative diseases

Martin Trapecar et al.

Summary: The research has developed a mesofluidic platform technology to study gut-liver-cerebral interactions, aiming to explore the pathophysiology of neurodegenerative diseases. Experimental results using a patient-derived Parkinson's disease brain model show that systemic interactions and microbiome-associated short-chain fatty acids are related to pathology-associated pathways in PD.
