RNA contributions to the form and function of biomolecular condensates

Christine Roden et al.

Summary: Recent studies have shown the significant contribution of RNA to cellular liquid-liquid phase separation and condensate formation, which play important roles in various cellular functions such as RNA transport, supporting catalytic processes, and stress responses. Although RNA and RNA-protein condensates are important in cells, the role of RNA in liquid-liquid phase separation has received less attention in comparison to RNA-binding proteins.Properties such as composition, length, structure, modifications, and expression levels of RNA can modulate the biophysical features of native condensates, influencing their size, shape, viscosity, and composition. RNA-protein condensates are involved in various cellular functions, including cell compartmentalization through RNA transport and localization, supporting catalytic processes, storage and inheritance of specific molecules, as well as buffering noise and responding to stress.

αα-Hub domains and intrinsically disordered proteins: A decisive combo

Katrine Bugge et al.

Summary: Hub proteins are essential in protein-protein interaction networks, with alpha alpha-hubs being a newly defined group of folded hub domains. They play a crucial role in organizing disordered transcriptional regulators and membrane scaffolds, showcasing multifaceted protein-protein interaction properties facilitated by the alpha alpha-hub fold.

Sequence-encoded and composition-dependent protein-RNA interactions control multiphasic condensate morphologies

Taranpreet Kaur et al.

Summary: The study highlights the importance of multivalent protein-protein and protein-RNA interactions in driving biological phase separation, demonstrating how competition between different molecules can lead to the demixing of condensates into stable coexisting phases. The biphasic condensates formed by a minimal system of PLP, RRP, and RNA exhibit diverse morphologies that are tunable through mixture stoichiometry and hierarchy of intermolecular interactions. The findings provide insights into the composition and spatial organization of multicomponent biomolecular condensates.

In vivo reconstitution finds multivalent RNA-RNA interactions as drivers of meshlike condensates

Weirui Ma et al.

Summary: Liquid-like condensates in cells are not necessarily sphere-like, as various condensates with filamentous morphology have been observed. One example is the TIS granule network, which shares a large surface area with the rough endoplasmic reticulum and plays a crucial role in membrane protein trafficking. Reconstitution experiments have shown that a multivalent RNA-binding protein and RNAs with large unstructured regions can form a pervasive intermolecular interaction network that acts as a condensate skeleton.
