Sonodynamic therapy for gliomas

Adomas Bunevicius et al.

Summary: Sonodynamic therapy (SDT) is a promising treatment approach for gliomas, involving the generation of reactive oxygen species and free radicals, as well as physical destabilization of cell membrane leading to cell death. Pre-clinical studies and animal models have shown improved tumor control and longer survival in animals treated with SDT. Clinical trials of SDT in patients with gliomas and other malignant brain tumors are currently underway.

Application of glutathione depletion in cancer therapy: Enhanced ROS-based therapy, ferroptosis, and chemotherapy

Boyi Niu et al.

Summary: Glutathione (GSH) plays a crucial role in cellular antioxidative system, particularly in cancer cells, where a high level of GSH is essential for scavenging ROS and detoxifying xenobiotics. Depletion of GSH has been shown to enhance the efficacy of various therapies such as ROS-based therapy, ferroptosis, and chemotherapy. This review comprehensively discusses the importance of GSH depletion in cancer therapy and various strategies employed to achieve it.

Hypoxia: The Cornerstone of Glioblastoma

Marta Domenech et al.

Summary: Glioblastoma is the most aggressive form of brain tumor in adults, with hypoxia-induced factor 1 (HIF-1) serving as an important driver of tumor progression by promoting angiogenesis, immunosuppression, and metabolic reprogramming. HIF-1 is not only regulated by oxygen levels but also by various oncogenic signaling pathways, and inhibiting the hypoxia pathway could be a crucial treatment alternative for GB patients.

Summary: Glioblastoma (GB) is the deadliest type of primary brain tumor with a median survival time of 16 months post-diagnosis, and radiotherapy remains the most effective treatment despite the common resistance observed in GB patients. Understanding the role of low tumor oxygenation (hypoxia) in radioresistance is crucial for developing novel strategies to improve tumor cell sensitivity to radiotherapy.

Increased DNA repair capacity augments resistance of glioblastoma cells to photodynamic therapy

Somayeh Shahmoradi Ghahe et al.

Summary: Resistance to photodynamic therapy in cancer cells may be caused by various mechanisms, including efficient repair of oxidative DNA damage and DNA breaks. Higher activity of APE1 endonuclease and increased expression and activation of DNA damage kinase ATM in the U-87 MGR cell line suggest that they are potential targets for sensitizing resistant cells to PDT.
