Impact of natural products on discovery of, and innovation in, crop protection compounds

Thomas C. Sparks et al.

Summary: Natural products (NPs) have been a significant source and inspiration for developing novel crop protection compounds, with NPs, their derivatives, and compounds inspired by NPs accounting for 17% of all crop protection compounds. The impact of NPs on the discovery of new modes of action and innovation in crop protection compounds has been consistent over the past 70 years. NPs have had the largest impact on insecticides compared to fungicides and herbicides.

Pharmaceutical technologies with potential application to insecticide discovery†

Keith D. Wing

Summary: New neuroactive insecticides are discovered/registered differently, with lower value in use compared to neuroactive pharmaceuticals, but show clear similarities in their biochemical modes of action. This paper examines recent trends in pharmaceutical science and identifies technologies that may represent complementary approaches to insecticide discovery and mode of action determination.

Status and outlook for acaricide and insecticide discovery

Peter Jeschke

Summary: The agrochemical industry is challenged by higher loss of active ingredients compared to the number being introduced, calling for the development of new products with improved efficacy and environmental profiles. Strategies include exploring acaricides and insecticides with new modes of action, or discovering novel molecules with activity against pest species with resistance-breaking properties.

Crop protection compounds - trends and perspective

Thomas C. Sparks et al.

Summary: The crop protection compound industry has undergone dramatic changes and consolidation since the late 1940s and early 1950s, with a decline in the introduction of new herbicides post-2000 being a major contributing factor to the overall perceived decline in numbers. However, excluding herbicides, the output of new fungicides and insecticides has remained relatively consistent, especially in the past 20 years. Innovation, measured by the number of compounds representing a new chemical class, has been relatively constant for the past 70 years, recently driven by new fungicides and insecticides.

Sulfoxaflor-A sulfoximine insecticide: Review and analysis of mode of action, resistance and cross-resistance

Gerald B. Watson et al.

Summary: Sulfoximines, represented by sulfoxaflor, are a relatively new class of insecticides that exhibit distinct interactions with metabolic enzymes and nicotinic acetylcholine receptors, resulting in minimal cross-resistance compared to other insecticides. Some insect strains show resistance to neonicotinoids but little to no cross-resistance to sulfoxaflor. In laboratory studies, high levels of resistance to sulfoxaflor have been observed, possibly linked to enhanced metabolism by specific cytochrome P450s.

East meets west: regional impact on agrochemical discovery and innovation

Thomas C. Sparks et al.

Summary: Efficient production of food is driving the demand for new crop protection compounds, with Europe, Japan, and the USA leading in their discovery and development. Over the past 75 years, there has been significant evolution in this process, characterized by consolidation and globalization among research and development-based companies. Despite a relatively constant rate of invention of new classes of crop protection compounds, there has been a shift towards Asia, particularly Japan, in introducing new compounds in the past 30 years.
