freqpcr: Estimation of population allele frequency using qPCR Delta Delta Cq measures from bulk samples

Masaaki Sudo et al.

Summary: PCR techniques are commonly used to estimate allele frequency in populations, with variation in DNA yields and measurement errors causing uncertainty in qPCR results. A statistical model was developed to estimate allele frequency and confidence intervals using Delta Delta Cq data from multiple bulk samples. The model showed unbiased estimates compared to simple averaging, with suggestions that dividing bulk samples for more precision or pooling into fewer samples for cost efficiency may yield comparable results.

Identification and characterization of striking multiple-insecticide resistance in a Tetranychus urticae field population from Greece

Kyriaki Maria Papapostolou et al.

Summary: A field population of Tetranychus urticae collected from an ornamental greenhouse in Peloponnese, Greece, exhibited resistance to multiple acaricides with multiple resistance mutations and over-expression of detoxification genes. Target-site resistance mutations against pyrethroids or bifenazate were not identified, possibly indicating reduced selection pressure on these chemistries in Greece.

Untangling a Gordian knot: the role of a GluCl3 I321T mutation in abamectin resistance in Tetranychus urticae

Wenxin Xue et al.

Summary: The study validated the functional role of the GluCl3 I321T mutation in abamectin resistance, with results showing a complex phenotypic strength. Using CRISPR-Cas9 mediated gene editing, it was demonstrated that the I321T mutation reduced the susceptibility of T. urticae to abamectin.

Resistance status and detoxification enzyme activity in ten populations of Panonychus citri (Acari: Tetranychidae) from Turkey

Ismail Doker et al.

Summary: Monitoring of acaricide resistance in phytophagous mites is crucial for effective pest management. A study conducted in citrus orchards in Cukurova region of Turkey found that certain populations of citrus red mites have developed resistance to commonly used acaricides. It is suggested that rotating acaricides with different modes of action is fundamental to prevent potential resistance problems.

Mechanisms and management of acaricide resistance for Tetranychus urticae in agroecosystems

Adekunle W. Adesanya et al.

Summary: The two-spotted spider mite poses a constant threat to sustainable production of economically important crops globally, with management heavily reliant on synthetic acaricides, which the mite has rapidly developed resistance to. Limited studies have been conducted on genetic changes related to acaricide resistance in T. urticae, necessitating further research for successful integrated mite management programs. Understanding the temporal and spatial characteristics of acaricide-resistant phenotypes and their underlying mechanisms is crucial for designing effective resistance management programs within various agroecosystems.

Summary: Tetranychus urticae Koch, a major agricultural pest, has rapidly developed resistance to pesticides, with field populations showing resistance to etoxazole and pyridaben and exhibiting cross-resistance. The detection of target site mutations in resistant populations highlights the importance of studying resistance patterns for designing sustainable IPM programs.
