Summary: Our study analyzed the Schistosoma mansoni SmKI-1 protein and designed two different point mutations on its Kunitz Domain, which were found to have enhanced NE inhibitory activity in vitro. In vivo data from the MSU-induced acute arthritis model demonstrated that the RL-KD mutant showed a notable anti-inflammatory effect compared to the EA-KD mutant. These results support the potential of parasitic helminth molecules as immunomodulatory agents for the development of novel therapies to control inflammatory diseases.
