Analysis of Stop Codons within Prokaryotic Protein-Coding Genes Suggests Frequent Readthrough Events

Summary: Nonsense mutations change a meaningful codon into a stop codon, resulting in a shortened protein product. In-frame stop codons are common in bacterial protein-coding genes but are not necessarily pseudogenes, as they are evolutionarily conserved. Double substitutions in codons show higher evolution rates, suggesting that in-frame stop codons can be introduced and reversed via positive selection.

Summary: The study identified a novel SXT/R391 ICE element, ICEPmiChnXH1653, carrying two tandem copies of bla(NDM-1) gene. The ICE was found to play a role in horizontal transfer of resistance genes and negatively impact bacterial fitness and transcriptome of the host. The research sheds light on the important role ICEs may play in the accumulation and transmission of antibiotic resistance genes.
