Genes from oxidative phosphorylation complexes II-V and two dual-function subunits of complex I are transcribed in Viscum album despite absence of the entire mitochondrial holo-complex I

Gitte Petersen et al.

Summary: Mistletoes such as Viscum have a unique electron transport chain with an almost complete absence of nuclear-encoded complex I genes, only expressing transcripts of two dual-function genes. Despite sequence divergence, the proteins encoded by these genes are structurally conserved. This complex I loss is a distinctive feature in Viscum species and their close relatives, making them a valuable system for studying the molecular consequences of its absence.

The gene space of European mistletoe (Viscum album)

Lucie Schroeder et al.

Summary: European mistletoe is a hemiparasitic plant with unique biochemical properties and an unusual cellular respiration process. Despite the large genome size of V. album, sequencing of its gene space has provided insights into its metabolism and molecular biology. The information from V. album gene space has led to the re-evaluation of mitochondrial proteome data and the identification of additional mitochondrial proteins.

Atomic structures of respiratory complex III2, complex IV, and supercomplex III2-IV from vascular plants

Maria Maldonado et al.

Summary: This study determined the atomic structures of mitochondrial complex III (CIII2), complex IV (CIV), and their supercomplex in Vigna radiata using single-particle cryoEM. The findings showed specific differences in plant complexes and challenged long-standing assumptions, providing new mechanistic hypotheses for proton translocation and complex interactions. The structures revealed coordinated movements and conformational heterogeneity within the complexes, offering insights into the unique features of plant respiration machinery.

Structure and assembly of the mammalian mitochondrial supercomplex CIII2CIV

Irene Vercellino et al.

Summary: The enzymes of the mitochondrial electron transport chain play a vital role in cell metabolism, forming supercomplexes in vivo. The assembly factor SCAF1 is crucial in mediating the assembly of CIII2CIV, but not involved in the assembly of the respirasome. Assembling into supercomplexes enhances the catalytic efficiency of CIII2 and CIV, potentially fine tuning the electron transfer efficiency in the electron transport chain.

Gamma carbonic anhydrases are subunits of the mitochondrial complex I of diatoms

Maximiliano Cainzos et al.

Summary: Diatoms are unicellular organisms with red algal-derived plastids, believed to have originated from endosymbioses with red algae or cryptophyta, with only chloroplasts remaining. Their mitochondria are thought to come from the exosymbiont, containing ancestral respiratory chains. Genes for gamma type carbonic anhydrases have been found in diatoms, suggesting an ancestral origin.

Summary: Through in vivo work and biochemical analysis, researchers found that a P25L mutation in the ND6 subunit of mitochondrial complex I provides protection against cardiac ischemia-reperfusion injury. This mutation does not affect oxidative phosphorylation but prevents complex I from generating ROS through RET, demonstrating the role of ROS in myocardial infarction.

Complexome profiling reveals novel insights into the composition and assembly of the mitochondrial ATP synthase of Arabidopsis thaliana

Helene Roehricht et al.

Summary: The mitochondrial ATP synthase, responsible for producing the energy needed for eucaryotic life, is structurally similar across species but exhibits some variations in plant-specific subunits. While assembly of this enzyme has been extensively studied in bacteria, yeast, and mammals, little is known about its assembly in plants. Through complexome profiling, researchers have identified assembly intermediates and proposed a model for the assembly pathway of plant mitochondria's ATP synthase, revealing that plant-specific subunits are also present in other organisms. This study demonstrates that the subunit composition and assembly pathway of plant mitochondria's ATP synthase are largely conserved with other mitochondrial enzymes.
