Disruption of NAP1 genes in Arabidopsis thaliana suppresses the fas1 mutant phenotype, enhances genome stability and changes chromatin compaction

Summary: The disruption of the H2A-H2B histone chaperone NUCLEOSOME ASSEMBLY PROTEIN 1 (NAP1) can suppress the loss-of-function phenotype of FAS1, leading to wild-type growth, decreased sensitivity to genotoxic stress, and suppression of telomere and 45S rDNA loss in Arabidopsis thaliana. This study demonstrates the essential role of NAP1 proteins in DNA repair in the absence of functional CAF-1, which is coupled to nucleosome assembly through modulation of H3 levels in the nucleus.

Cytoskeleton integrity influences XRCC1 and PCNA dynamics at DNA damage

Summary: DNA damage induced by 405-nm laser light recruits proteins involved in base excision repair (BER), but perturbing actin or tubulin polymerization in human cells alters the dynamics of BER factors. Depolymerization of cytoplasmic actin may compromise BER efficiency in mammals due to increased levels of nuclear actin and tubulin, linking cytoskeletal integrity to BER.

Super-resolution mapping of cellular double-strand break resection complexes during homologous recombination

Summary: Using single-molecule localization super-resolution imaging assays, we observed the spatiotemporal behavior of key mediator and nuclease proteins during DNA resection at single-ended double-strand breaks. Multiple simultaneous resection events were demonstrated, with recruitment of various proteins and completion of resection 2 to 4 hours after break induction. Additionally, we identified potential roles of BRCA1 and BLM in homology search and repair resolution during HR.

Impact of chromatin context on Cas9-induced DNA double-strand break repair pathway balance

Summary: Through systematically analyzing the impact of chromatin context on three DNA double-strand break repair pathways, it was found that non-homologous end-joining is more common in euchromatin, while microhomology-mediated end-joining is more active in specific heterochromatin contexts. Additionally, single-stranded template repair competes strongly with other repair pathways and is moderately linked to chromatin context.

Gene dosage compensation of rRNA transcript levels in Arabidopsis thaliana lines with reduced ribosomal gene copy number

Summary: In this study, the researchers used Cas9-mediated genome editing in Arabidopsis to reduce the copy number of 45S rRNA genes to around 10%, creating low copy number (LCN) lines. Despite the significant reduction in rDNA copies, the transcriptional rates and steady-state levels of rRNA remained the same as wild-type plants. Furthermore, editing rRNA repeats using Cas9 technology provides insights into the mechanisms of rDNA dosage compensation and the impacts of low rDNA copy number on genome stability, development, and cellular processes.
