ACTA2-Related Dysgyria: An Under-Recognized Malformation of Cortical Development

S. Subramanian et al.

Summary: Pathogenic variants in the ACTA2 gene are associated with a distinctive arterial phenotype and gyral abnormalities in the brain. Gyral abnormalities, commonly found in patients with ACTA2 pathogenic variants, are predominantly located around the Sylvian fissure and in the frontal lobe, often co-occurring with the characteristic ACTA2 arterial phenotype.

ACTA2 mutation is responsible for multisystemic smooth muscle dysfunction syndrome with seizures: A case report and review of literature

Wen-Xian Yang et al.

Summary: The ACTA2 gene encodes actin alpha 2 and mutations in this gene can cause Multisystem smooth muscle dysfunction syndrome (MSMDS), which is characterized by aortic and cerebrovascular lesions. This case report highlights a 7-year-old girl with recurrent cough, asthma, and seizures, who was diagnosed with MSMDS and epilepsy after genetic testing.

Cerebral arteriopathy in ACTA2 mutations: a spectrum of disease highlighted by a case of variable penetrance in two siblings

Christine Tschoe et al.

Summary: Cerebral arteriopathy caused by heterozygous mutations in the ACTA2 gene is now recognized as a distinct cerebrovascular entity with unique angiographic features. The penetrance and systemic manifestations of these mutations vary based on the location of the amino acid replacement within the smooth muscle actin protein. Further research is needed to assess the safety and efficacy of revascularization procedures for this condition, and early recognition and genetic testing are crucial in determining clinical outcomes.
