The annual incidence rate of PsA was significantly lower in the bDMARDs group compared to the phototherapy group. Factors associated with a higher risk of PsA included older age, nail psoriasis, and psoriasis duration over 10 years.

Circulating Mir-140 and leptin improve the accuracy of the differential diagnosis between psoriatic arthritis and rheumatoid arthritis: a case-control study

Summary: MicroRNAs and adipokines were investigated as potential additional markers to differentiate psoriatic arthritis (PsA) from rheumatoid arthritis (RA). The study found that the gene expression of miR-140 was up-regulated in PsA patients and down-regulated in RA patients compared to healthy controls, and it was significantly higher in PsA compared to RA. Serum levels of IL-23a and leptin were significantly increased in both PsA and RA patients compared to healthy controls. These findings suggest that circulating miR-140 and serum leptin could be potential additional biomarkers for discriminating PsA from RA.

Summary: This study investigated the potential of the human gut microbiome in predicting methotrexate (MTX) efficacy in new-onset rheumatoid arthritis (RA) patients. The analysis revealed significant associations between gut bacterial taxa abundance and genes with future clinical response, as well as a microbiome-based model that predicted lack of response to MTX. Ex vivo experiments also suggested a possible direct effect of the gut microbiome on MTX metabolism and treatment outcomes, supporting the value of the gut microbiome as a potential prognostic tool and therapeutic target in RA.

Ex vivo mass cytometry analysis reveals a profound myeloid proinflammatory signature in psoriatic arthritis synovial fluid

Summary: By using mass cytometry combined with transcriptomic analysis, this study identified significant differences in myeloid cell populations between psoriatic arthritis (PsA) synovial fluid (SF) and blood, with expansion of intermediate monocytes, macrophages, and dendritic cells in SF. Myeloid cells, particularly classical monocytes, intermediate monocytes, and macrophages, were found to produce proinflammatory mediators like osteopontin and CCL2 spontaneously in PsA SF, while minimal cytokine production was detected in T cells. Gene expression analysis revealed upregulation of osteopontin and CCL2 genes in PsA monocytes/macrophages, which were also elevated at the protein level in PsA SF.
