Preparation and Evaluation of Novel Supersaturated Solid Dispersion of Magnolol

Summary: This article aimed to design a new type of supersaturated solid dispersion (NS-SD) loaded with Magnolol (Mag) to increase oral bioavailability in rats. The optimum carrier, Plasdone-630 (PS-630), was selected based on solubility parameters, phase solubility experiments, inhibition precipitation experiment, and in vitro release experiment. By adding Monoglyceride (MG) and Lecithin High Potency (LHP) into the Mag-S-SD, the dosage of carrier was reduced and the release rate was improved. The prepared Mag-NS-SD showed increased bioavailability in vivo compared to Mag and Mag-S-SD.

Summary: This study prepared supersaturable self-emulsifying drug delivery systems (QT-sSEDDS) loaded with quercetin (QT) and evaluated their characteristics. The results demonstrated that QT-sSEDDS exhibited superior solubilization potential and enhanced cell uptake compared to conventional QT-SEDDS. The in vivo pharmacokinetic data further confirmed the increased drug concentration and bioavailability of QT-sSEDDS.

Molecular metadynamics simulations indicated that CC interacts more strongly with HPMCAS than HCTZ.
