Recent Design and Structure-Activity Relationship Studies on the Modifications of DHFR Inhibitors as Anticancer Agents

Agnieszka Wrobel et al.

Summary: This review summarizes recent advances in the research of new DHFR inhibitors with potential anticancer activity over the past decade. It highlights the importance of thorough physicochemical characterization and biological investigations in understanding the structure-activity relationship of DHFR inhibitors for better design and synthesis of active compounds. Multiple studies have been conducted on the modification of known DHFR inhibitors and the design of new molecules as potential anti-cancer drugs.

Design, Synthesis, Docking and Biological Evaluation of Novel 4-hydroxy Coumarin Derivatives

N. Ramalakshmi et al.

Summary: This study focused on designing 36 novel coumarin derivatives with potential antibacterial activity against quinolone-resistant E. coli strains. Docking studies showed that compounds with electron-withdrawing groups exhibited increased antibacterial activity. Compounds 6, 23, and 30 demonstrated higher activity in MIC results, suggesting the need for further in vivo evaluation.

Molecularly targeted therapy for advanced gastrointestinal noncolorectal cancer treatment: how to choose? Past, present, future

Olga Nigro et al.

Summary: Gastrointestinal cancer is a major cause of death globally, with conventional cytotoxic chemotherapy being a key element in treatment but showing limited improvements in survival outcomes. Recent progress in molecular biology of noncolorectal gastrointestinal cancers has led to the discovery of specific targets and development of systemic drugs blocking critical kinases and molecular pathways. New clinical trials are incorporating classic chemotherapy with small-molecule inhibitors or mAbs to further improve treatment outcomes.
