PROTAC targeted protein degraders: the past is prologue

Miklos Bekes et al.

Summary: Targeted protein degradation (TPD) is a new therapeutic modality that can tackle disease-causing proteins which are difficult to target with conventional small molecules. PROTAC molecules, utilizing the ubiquitin-proteasome system to degrade target proteins, has achieved clinical proof-of-concept and attracted significant industry activity. Future directions include identifying target classes suitable for TPD, expanding the use of ubiquitin ligases for precision medicine, and extending the modality beyond oncology.

Inclusion complexation of the anticancer drug pomalidomide with cyclodextrins: fast dissolution and improved solubility

Zoltan-Istvan Szabo et al.

Summary: The inclusion complexation of pomalidomide with sulfobutylether-beta-cyclodextrin significantly enhances the water solubility of pomalidomide and maintains its anti-angiogenic activity. Various analytical techniques were employed to characterize the complex, confirming a 1:1 stoichiometry between pomalidomide and sulfobutylether-beta-cyclodextrin.

Enteric reabsorption processes and their impact on drug pharmacokinetics

Manuel Ibarra et al.

Summary: Enteric reabsorption refers to the process in which a drug is secreted into the intestinal lumen and reabsorbed into the systemic circulation, primarily involving mechanisms such as hepatobiliary drug secretion and gastric secretion. The impact of drug reabsorption on systemic clearance, volume of distribution, and bioavailability depends on the type of drug and hepatic extraction. Increases in enterohepatic reabsorption can significantly affect drug clearance, bioavailability, and volume of distribution, while enterogastric reabsorption has negligible impact on these pharmacokinetic parameters.

Bioanalysis of niclosamide in plasma using liquid chromatography-tandem mass and application to pharmacokinetics in rats and dogs

Hae-In Choi et al.

Summary: A study developed a LC-MS/MS method for determining niclosamide concentration in animals, revealing its antiviral effects against the SARS-CoV-2 virus. Pharmacokinetic studies showed dose-independent pharmacokinetics of niclosamide in rats and dogs, with low drug exposure after oral administration.
