Inhibition of Viral Replication Reduces Transcriptionally Active Distinct Hepatitis B Virus Integrations With Implications on Host Gene Dysregulation

Yao-Chun Hsu et al.

Summary: This study aimed to assess the effect of viral inhibition on hepatocellular carcinogenesis of hepatitis B virus (HBV) infection and explore the correlation between viral integrations and host gene dysregulation. The results showed that viral inhibition can reduce the number of transcriptionally active viral integrations and viral integrations are significantly correlated with host gene dysregulation.

Recent Progress and Future Prospective in HBV Cure by CRISPR/Cas

Yu-Chan Yang et al.

Summary: Hepatitis B virus (HBV) infection remains a global public health issue, and the current antiviral therapy fails to cure it. CRISPR-mediated gene editing has emerged as a promising approach for HBV cure, but it faces challenges such as off-target effects and difficulty in finding conserved target sequences.

Ongoing viral replication and production of infectious virus in patients with chronic hepatitis B virus suppressed below the limit of quantitation on long-term nucleos(t)ide therapy

Dara L. Burdette et al.

Summary: Nucleos(t)ide analogs are standard treatment for chronic hepatitis B but do not eliminate the viral episome, leading to the presence of residual infectious virus in some treated patients. Additional treatment intensification may be necessary to achieve a cure.

Mitochondrial targeted meganuclease as a platform to eliminate mutant mtDNA in vivo

Ugne Zekonyte et al.

Summary: This study shows the potential of using mitoARCUS, a mitochondrial-targeted gene editing platform, to specifically eliminate mutant mtDNA with a m.5024C>T mutation in the mt-tRNA(Ala) gene. MitoARCUS was successful in robustly eliminating mutant mtDNA in the liver and skeletal muscle of mice, demonstrating its promise as a powerful tool for the treatment of heteroplasmic mitochondrial DNA mutations.

Inhibition of Hepatitis B Virus by AAV8-Derived CRISPR/SaCas9 Expressed From Liver-Specific Promoters

Kun Yan et al.

Summary: Genome editing technology CRISPR/Cas may hold potential in treating chronic hepatitis B virus infection, especially when combined with liver-specific promoters to effectively inhibit HBV replication, while ensuring safety is essential. The study results are helpful for restricting the effects of the CRISPR/Cas9 system to the liver in clinical applications, thereby reducing the probability of nonspecific targeting in other organs.
