Depletion of Trp53 and Cdkn2a Does Not Promote Self-Renewal in the Mammary Gland but Amplifies Proliferation Induced by TNF-α

Linda J. van Weele et al.

Summary: The mammary epithelium undergoes extensive proliferation during the female reproductive cycle, regulated by mammary stem cells and tumor suppressor proteins. While p53, p16(INK4a), and p19(ARF) are not significant in limiting normal mammary epithelium self-renewal and proliferation, they may play a role in preventing excessive proliferation in the presence of inflammatory cytokines like TNF-alpha.

Transgenic mouse models of breast cancer

Angelina T. Regua et al.

Summary: Transgenic breast cancer mouse models are essential for preclinical studies, allowing for modeling of abnormal genetic events commonly seen in human breast cancers. By employing tissue-specific genetic manipulation, these models can mimic spontaneous mammary tumorigenesis by expressing oncogenes or deleting tumor suppressors. The variability in characteristics and strain-dependent features of these models can be utilized to study different aspects of mammary tumorigenesis and metastasis.

Acquired Resistance to KRASG12C Inhibition in Cancer

M. M. Awad et al.

Summary: A study of 38 patients with KRAS(G12C)-mutant cancers treated with adagrasib revealed diverse mechanisms of acquired resistance in 45% of them, including various mutations and bypass mechanisms. New therapeutic strategies are needed to overcome this drug resistance.
