The importance of immune checkpoints in immune monitoring: A future paradigm shift in the treatment of cancer

Hajar Alemohammad et al.

Summary: The growth and development of cancer are directly correlated to the suppression of the immune system, with immune checkpoints playing a key role in inhibiting anti-tumor immune responses. Up-regulation of inhibitory immune checkpoints on immune cells during tumor progression suppresses anti-tumor immune responses and promotes immune escape. Targeting inhibitory immune checkpoints through antibodies or miRNAs is a promising therapeutic strategy, and immune checkpoint inhibitors have shown favorable results in enhancing immune cell-induced antitumor responses.

Tumor-selective blockade of CD47 signaling with a CD47/PD-L1 bispecific antibody for enhanced anti-tumor activity and limited toxicity

Yan Wang et al.

Summary: The newly designed CD47/PD-L1 bispecific antibody, IBI322, effectively inhibits CD47-SIRP alpha signal, promotes tumor cell phagocytosis, accumulates in PD-L1-positive tumors, shows synergistic therapeutic effects, and has minimal impact on red blood cells, making it a promising option for cancer treatment.

Targeting CD47 for cancer immunotherapy

Zhongxing Jiang et al.

Summary: Significant progress has been made in targeting CD47 for cancer immunotherapy in solid tumors and hematological malignancies, with a total of 46 clinical trials related to CD47 in the USA and 23 in China. Most of the trials are in early phases, and the new generation CD47 antibodies have shown promising results.

The role of CD47-SIRPα immune checkpoint in tumor immune evasion and innate immunotherapy

Zihao Li et al.

Summary: CD47, as a transmembrane protein, plays a crucial role in modulating immune evasion of tumor cells. In recent years, CD47-SIRP alpha immune checkpoint inhibitors have gained increasing attention for their potential in tumor immunotherapy. Understanding the function of CD47 in the tumor microenvironment has accelerated the development of treatment strategies targeting this immune checkpoint.
