Garciesculenxanthone B induces PINK1-Parkin-mediated mitophagy and prevents ischemia-reperfusion brain injury in mice

Man Wu et al.

Summary: Mitophagy is a selective form of autophagy that removes damaged mitochondria via the autophagy-lysosome pathway. GeB, a natural compound, can promote the PINK1-Parkin-mediated mitophagy pathway, potentially protecting against I/R brain injury.

Mitochondrial dysfunction in cardiovascular disease: Current status of translational research/clinical and therapeutic implications

Antonis S. Manolis et al.

Summary: Mitochondria play a crucial role in providing energy for cells and have various functions beyond energy production. Dysfunction of mitochondria can lead to cardiomyopathies and other cardiovascular diseases, often caused by mutations in mitochondrial or nuclear DNA, as well as aging, disease, and environmental stresses. Developing targeted therapeutic strategies for mitochondria remains a challenge due to variable responses based on the underlying causes of dysfunction.

Netrin-1 and its receptor DCC modulate survival and death of dopamine neurons and Parkinson's disease features

Melissa Jasmin et al.

Summary: The netrin-1/DCC ligand/receptor pair plays key roles in CNS development, and although they are expressed in the adult brain, their function in mature neurons is not well understood. Research shows that netrin-1 plays a critical role in the fate of adult dopamine neurons, and targeting netrin-1 signaling may have therapeutic potential in Parkinson's disease. Studies in animal models suggest that overexpression of netrin-1 or administration of recombinant netrin-1 can be neuroprotective and neurorestorative in PD.

Mitochondrial dysfunction in the development and progression of neurodegenerative diseases

Joseph Johnson et al.

Summary: Mitochondria play crucial roles in modulating apoptosis, ferroptosis, and inflammasome activation, in addition to ATP synthesis. Dysfunction in these organelles contributes to the development and progression of neurodegenerative diseases, making targeting mitochondrial dysfunction a potential therapeutic strategy. Therapeutics are being developed to circumvent dysfunctional mitochondria's roles in signaling for inflammasome activation and ferroptosis.
