Novel amide derivatives of 3-phenylglutaric acid as potent soluble epoxide hydrolase inhibitors

Summary: The study focused on designing, synthesizing and evaluating new amide-based derivatives of 3-phenylglutaric acid as potential sEH inhibitors. Most of the synthesized compounds showed nanomolar range inhibition against sEH enzyme, with the best observed IC50 value of 0.5 nM. Incorporating carboxylic moieties into the structures could increase solubility and improve physicochemical properties.

The EH family comprises 5 proteins and 2 candidate members, with the best-known EHs being EPHX1 and EPHX2, which have wide expression patterns and multiple functions, leading to the development of specific inhibitors. Studies on pathogenicity of variants in EH genes are ongoing, highlighting the pleiotropic role of EHs in various disorders.

Design, synthesis, and biological evaluation of some 1,2,3-triazole derivatives as novel amide-based inhibitors of soluble epoxide hydrolase

Summary: In this study, a series of novel amide derivatives with 1,2,3-triazole ring as a secondary pharmacophore against sEH enzyme were designed and investigated. Most synthesized compounds showed satisfactory inhibitory activity against sEH, with compound 5l exhibiting the most potent IC50 value of 0.9 nM. In silico ADME calculations indicated that these compounds could potentially be formulated as orally active agents.
