Tenascin-C preserves microglia surveillance and restricts leukocyte and, more specifically, T cell infiltration of the ischemic brain

Daniel Manrique-Castano et al.

Summary: The study shows that TnC deficiency affects the morphology and quantity of microglial cells, reducing their surveillance capabilities and increasing the accumulation of infiltrating leukocytes (especially T cells) in the ischemic brain.

Stroke subtype-dependent synapse elimination by reactive gliosis in mice

Xiaojing Shi et al.

Summary: This study showed that inhibiting phagocytosis of microglia and astrocytes in ischemic stroke improved neurobehavioral outcomes, attenuated brain damage, and prevented synapse loss. In hemorrhagic stroke, inhibiting phagocytosis of microglia/macrophages but not astrocytes improved neurobehavioral outcomes. Single-cell RNA sequencing revealed downregulation of phagocytosis-related processes in astrocytes of the hemorrhagic brain compared to the ischemic brain.

Knock-Out of Tenascin-C Ameliorates Ischemia-Induced Rod-Photoreceptor Degeneration and Retinal Dysfunction

Susanne Wiemann et al.

Summary: This study aimed to investigate the functional role of the glycoprotein tenascin-C (Tnc) in retinal ischemia, finding increased Tnc levels after ischemia accompanied by diminished rod-photoreceptor degeneration and retinal dysfunction in KO mice. Additionally, KO mice showed lower levels of activated caspase 3(+) cells, suggesting a potential role of Tnc in synaptic remodeling during ischemic retinal degeneration. Increased vesicular glutamate transporter 1 (vGlut1) levels in both ischemic groups indicated impaired glutamatergic signaling and excitotoxicity after ischemia.
