Identification of structurally diverse menaquinone-binding antibiotics with in vivo activity against multidrug-resistant pathogens

Summary: Meta-omic techniques were used to identify new menaquinone-binding antibiotics with activities against multidrug-resistant bacteria. The discovered antibiotics are structurally diverse and show potential for treating methicillin-resistant Staphylococcus aureus and Mycobacterium tuberculosis infections. The study highlights the importance of exploring naturally occurring antibiotics for the development of new antibiotics with different mechanisms of action.

N-acyl-L-homoserine lactones release and microbial community changes in response to operation temperature in an anammox biofilm reactor

Summary: The study found that the decrease in temperature weakened quorum sensing and led to a reduction in nitrogen removal efficiency in the anaerobic ammonium oxidation reactor. Candidatus Kuenenia was identified as the predominant anammox bacteria, with its abundance increasing as the temperature decreased. The complex relationship between microbial genus and AHLs was also observed.

Novel Macrolactams from a Deep-Sea-Derived Streptomyces Species

Summary: Four polyene macrolactams, including three new ones, were isolated from the fermentation broth of a deep-sea sediment-derived Streptomyces sp. Their structures were determined using spectroscopic analysis and X-ray diffraction. Two of the compounds showed selective antifungal activity against Candida albicans.

Chemical Elicitors Induce Rare Bioactive Secondary Metabolites in Deep-Sea Bacteria under Laboratory Conditions

Summary: The study describes an integrated methodology for the construction and screening of an elicited and pre-fractionated library of marine bacteria, using chemical elicitors to induce the expression of cryptic biosynthetic gene clusters. The results show that elicited bacterial metabolites correspond to around 45% of compounds produced under laboratory conditions, with a novel chemical space of around 70% or concentrated in the chemical space of drugs.

Natural polyenic macrolactams and polycyclic derivatives generated by transannular pericyclic reactions: optimized biogenesis challenging chemical synthesis

Summary: Genetically-encoded polyenic macrolactams, constructed by Nature using hybrid polyketide synthase/nonribosomal peptide synthase assembly lines, are a large collection of natural products isolated from bacteria. Recent activation of cryptic gene clusters in microorganisms allows for the generation and isolation of additional members, resulting in structurally diverse polycyclic products and oxidized derivatives through transannular reactions and rearrangements. Scientists face challenges in isolating these unstable compounds from cultures, characterizing their structures, determining biological activities, optimizing biogenetic pathways, and exploring skeletal rearrangements derived from pericyclic reactions of the macrolactams. Synthetic chemists are also making efforts to emulate Nature in successfully generating and confirming the structures of these natural products.
